Zanthoxylum dissitum Hemsl belonging to the Rutaceae Zanthoxylum Hemsl, is one of the important valuable and rare woody medicinal plant, all its root, stem and leaves, which could activate blood circulation to dissipate blood stasis, resolve toxin and disperse swelling. Mainly used for gynecological medicine manufacturing. In recent years, because of domestic production scale, and Zanthoxylum dissitum Hemsl seed natural germination rate is low, and the growth speed is slow, resulting in Zanthoxylum dissitum Hemsl natural resources is poor, can not meet the demand of the related pharmaceutical factory. In this research, we do artificial germination of Zanthoxylum dissitum Hemsl seeds, Then examine the endogenous hormone and nutrient content and enzyme activities during the germination of seeds of Zanthoxylum dissitum Hemsl seeds. The results are as follows:1, Artificial germination of seeds of Zanthoxylum dissitum HemslThrough single factor and orthogonal test, the optimum scheme of seed germination of Zanthoxylum dissitum Hemsl is:soaking the seed with80%concentrated sulfuric acid for3min; turn to the sodium bicarbonate solution (30g/L) soaking for10min; after broken the shell of seeds, soaking with GA3(540mg/L)for5.5h; Finally turn to the nutrient solution with6-BA (2.7mg/L) and CaCl2(5g/L).Using this project, the germination rate of Zanthoxylum dissitwn Hemsl seeds can reach to83%. In this research, seeds began to germinate in third days, in the fourth day, in the fourth day, germination rate will reach maximum. And the hypocotyls will elongation fast in the next2-3days, in the seventh day to the eighth day part of cotyledon began to gradually become pale green.2,Changes of endogenous hormone during the germination of the seeds of Zanthoxylum dissitwn HemslThis study was analyzed the endogenous hormones in seeds of Zanthoxylum dissitum Hemsl by high performance liquid chromatography method. Mobile phase using methanol, acetonitrile,1%acetic acid(30:5:65); select wavelength254nm; Flow velocity choose0.5ml/min.Soaking with GA3, the endogenous hormones content of seeds will become less, But after began the germination experiment, the endogenous hormones content of experimental group seeds will rose sharply in early stage. This phenomenon is maybe related with Hormonal regulation of seed germination, and the growth of the seeds hypocotyls. After rising, hormone cuntent began to drop, to sixth days began to slowly rise again, This phenomenon may be due to hormone has been completed germination regulation. While the hormone content rise again after sixth days may be related with embryo differentiation.3, Changes of nutriment and enzyme activity during the germination of the seeds of Zanthoxylum dissitwn Hemsl。This proof the starch and protein has been decomposed in the process of seeds germination, and release of energy, and generate a large amount of soluble sugar. But declined in sixth days of soluble content, which is related to the Embryo growth and the starch, protein content and amylase activity decreased. While the content of soluble sugar in seventh days and began to rise again, this should be with the hydrolysis of fat related. The fat content is from the beginning of the sixth day there has been a significant downward trend.a-amylase and β-amylase catalyze78.6%of starch Hydrolyze to Small molecular sugar, that provide carbon nutrition and energy for seed germination and growth stage, ensures substance and energy supply in the process of seeds germination...
