Aquilegia viridiflora pall is perennial herbs of Ranunculaceae in Aquilegia, the whole plant can be used as medicine which are medicine and edible plants of the Mongolian folk. Generally, from May to June we can collect their spear leaves as edible wild vegetables, there are rich in nutrients. From July to August we can pick the whole plant as into medicine, They have many function, for example regulate menstruation, detoxification, anti-inflammatory, analgesic. Aquilegia viridiflora widely distributed in the Inner Mongolia region, has a long history in folk use, easy to pick, easy to identify. This study was analyzed active ingredient content and structure identification of alkaloids.In this study, use the Mongolian medicinal and edible plants Aquilegia viridiflora as materials, select different picking periods to detect the Alkaloids、flavonoids、vitamin C content、protein content and choose different growth stages to detect the total antioxidant capacity (T-AOC), glutathione peroxidase (GSH-PX) content, catalase (CAT) content and total superoxide dismutase (T-SOD) content and the dynamic changes of the above analysis; Determination of the ethanol extract of Aquilegia that can or can not affect the Drosophila life span, body CAT、SOD and malondialdehyde (MDA) content; By using orthogonal experiment to determined the optimum conditions of total alkaloids; By using efficient gas chromatography-mass spectrometry (GC-MS) combined with technology, total alkaloids of Aquilegia viridiflora were isolated and purified and Identification。Through this study, enrich and improve Aquilegia effective component research to provide the scientific basis for its clinical application, Let Aquilegia viridiflora play a greater value, reveal the seasonal variation of the active ingredient, for a reasonable harvest, protection of national medicinal and edible plant resources provide scientific ideas and experimental data.The results are as follows;1 Dynamic study of Aquilegia viridiflora metabolite contentAquilegia viridiflora picking time for 2014 of May 13, June 5, June 28, July 21, August 13。By experiment, results showed that Aquilegia viridiflora alkaloid content was highest at August 13; The flavonoid content was highest at June 28; The protein content was highest at July 21; The vitamin C was highest at June 5; T-AOC content was highest at August 13; T-SOD content was highest at June 28; CAT content was highest at August 13; GSH-PX content was highest at July 21。2 Study of Aquilegia viridiflora antioxidant activityWhen using (0.06 mg/100 mg) concentration Aquilegia viridiflora ethanol extract fed female and male Drosophila and that can extend the life of 10 days and 3 days, the average extension life rate of male and female Drosophila is 16.32% and 5.36%; When using (0.12 mg/100 mg) concentration Aquilegia viridiflora ethanol extract fed female and male Drosophila and that can extend the life of 11 days and 4 days, the average extension life rate of male and female Drosophila is 18.35% and 5.84%。 Aquilegia viridiflora various concentrations of ethanol extract can improve male and female Drosophila CAT, SOD activity and reduce the content of MDA。3 Study of the extraction crafts of alkaloids Aquilegia viridifloraUsing ethanol as solvent to extract Aquilegia viridiflora total alkaloids, For four main factors that is the ethanol concentration、solid-liquid ratio、 extraction time and extraction temperature, design single-factor experiment and obtain optimal levels of each factor, and than design L9 (34) orthogonal experiment. The study found four factors that affect the order of Aquilegia viridiflora total alkaloid extraction rate is B=D> A> C, that extraction temperature=ethanol concentration>extraction time> solid-liquid ratio, experimental optimum combination B2D2A2C2, that extraction temperature was 70℃, ethanol concentration was 80%, extraction time was 60min, solid-liquid ratio was 1:25.4 Purification and identification of Aquilegia viridiflora total alkaloidAquilegia viridiflora ethanol extract and ethanol extract of ultrasonic-assisted alkaloids both have qualitative precipitation reaction, ethanol extract of ultrasound-assisted qualitative response to modified bismuth potassium iodide reagents and reagent Frobde than ethanol extract significantly. It is description that ethanol extract and ethanol extract of ultrasound-assisted both contain alkaloids, and ultrasound-assisted extraction has more advantages than pure ethanol solvent extraction. Finally, we separate two kinds of alkaloids, they are both Schiff base compounds as 2,6-Dimethoxybenzaldehyde carbamoylhydrazone and 2-(3-Chlorophenoxy)-N’-(2,4-dimethoxybenzylidene)acethydrazide.In conclusion, through the Aquilegia viridiflora basic research, established best edible and medicinal picking time. The results indicated if we want to eat Aquilegia viridiflora, we should choose the low alkaloid content and high protein and vitamin C content, that is June. If we want to use as medicine Aquilegia viridiflora, we should choose higher alkaloid content in August. Aquilegia viridiflora ethanol extracts of Drosophila has anti-aging effects of life extension. By using orthogonal experiment to determined the optimum conditions of total alkaloids that extraction temperature was 70℃, ethanol concentration was 80%, extraction time was 60min, solid-liquid ratio was 1:25. we separated two kinds of alkaloids. The above results for Aquilegia active ingredient research and rational development and utilization were provided experimental data.
