To investigate the prevalence of Brucellosis in the dairy farms in Corps Twelfth Division in Xinjiang, 3531 serum samples, 20 diseased tissue samples and 20 milk samples were collected from 5 large‐scale dairy farms and farming community located in A farm, B farm, C farm, D farm and E farm. Rose bengal plate agglutination test(RBPT), slow agglutination test(SAT), AMOS‐PCR and VirB8‐PCR were applied in investigating the epidemiology of brucellosis. The RBPT and SAT test results showed that the positive rates of brucellosis were 13.4%、0、5.1%、0.87%、and 0.51% in cows in A Mission, B farm, C farm, D farm and E farm, respectively, while 5.3%、4.7% and 0 in large‐scale dairy farms, farming community and backyard farmers dairy herds, respectively. PCR was applied on typing the Brucella strains from milk of brucellosis positive cows. The result showed that the Brucella in farms were mainly type3, 5, 5, and 9, while A19 vaccine presented type I Brucella biological characteristics, which had a strong guiding significance for the region on brucellosis prevention and control.A part of the adult cows and calves were inoculated with S2 and A19 live vaccine for brucellosis based on the epidemiological investigation. The results indicated that the positive immune antibodies were 31% 30 days after inoculation of S2 live vaccine, while 100% 20 days after A19 live vaccine inoculation. Positive rate for brucellosis in cows after subcutaneous inoculation with A19 was higher than that for oral inoculation with S2. Inoculation with live vaccine in cows can effectively control brucellosis. Results from SAT testing before and after inoculation and results from comprehensive prevention and control showed that related diseases, including abortion rate in pregnant cattle, endometritis, mastitis, arthritis, afterbirth, empty infertility pregnant, were decreased 18.5%、8.8%、7.5%、3.3%、7.2%、5.9%,respectively.Based on the investigation of brucellosis epidemiology and the monitoring of immunological effect in dairy farming community in twelfth division, inoculation procedures, quarantine standards, technical specifications harmless treatment disinfection, disposal technologies and standardized quarantine system were developed and applied in the dairy farming district. These measures ha made effective effects on prevention and control of cow brucellosis in dairy farming community in the twelfth division.
