| In this paper, total alkaloids of Sophora alopecuroides(TASA) were investigated which were manufactured into water-soluble perfusion agent and establish quality control standard. The optimal parameters of extraction were obtained by single factor experiments and orthogonal tests with the total alkaloids were determined by acid dye colorimetry. Through the effects of Sophora alopecuroides(AL) on acute toxicity in mice and antibacterial experiment in vitro to provide the basis for pharmic content. The quantitative determination and qualitative determination of Sophora alopecuroides perfusate were measured Respectively by high performance liquid chromatography(HPLC) and thin layer chromatography(TLC). To establish quality standard of Sophora alopecuroides perfusate. The optimal formulation of extraction technology was selected by L9(34) orthogonal design. The optimal moulding process is 1:20 of solid-liquid ratio, p H 0.5 water/ hydrochloric acid and extracting two times and 18 h one time. The contents are 7.08% which determined by acid dye colorimetry. Eventually, the total alkaloid of sophora alopecuroides perfusion agent effective dose is 120 mg/m L. The sophoridine is identified by thin layer chromate- graphy(TLC) and determined by high performance liquid chromatography(HPLC).Aqueous solution of total alkaloid of sophora alopecuroides are physically and chemically stable and has a good ability of diffusion. After injected into the uterus, it can avoid local stimulation for high concentration and clean the endometrium. It was proved that the preparation techniques of total alkaloid of sophora alopecuroides perfusion agent is reasonable and practical, the quality of preparation is simple and controllable. Total alkaloid of sophora alopecuroides is a natural product. So it will not have developed a resistance and drug residues. It has good antibacterial and anti-inflammatory effects. Nowadays socity, abusing of antibiotics is becoming increasingly which provided a broad research potential and application prospect for TASA. |
