In this study, we discussed the mechanisms of SP modulating β-catenin and BMP2/4and its possible signal pathway.First we discussed the most suitable concentration of SP for skin stem cells. We set up5different concentrations of1×10’5mol/L and1X10-6mol/L,1X10"7mol/L,1X10-8mol/L,1X10"9mol/L. After stimulated with different concentrations of SP, we made its growth curves and examined expressions of β-catenin and BMP2/4by using quantitative PCR at different day. The results indicated that1X10"6mol/L is the most suitable concentration of SP for this study. We also observed the phenomenon that β-catenin and BMP both at high level of gene expression.Previous research shows that SP has three types of receptor, NK1-R and NK2-R, NK3-R. Atfer NK1-R, NK2-R, both NK1-R and NK2-R of cultured skin stem cells in vitro were blocked respectively and stimulated by1×10-6mol/L SP, the expression of β-catenin and BMP2/4were examined by quantitative PCR. Results shows that SP regulate β-catenin mainly by NK2-R. and in the process of SP regulate BMPs, possibly NK3-R play an important role. Also there is probability that unknown approach work in the process of SP regulate BMPs.
