In my studies on the germplasm resource in cultivated strains of Tricholoma giganteum massee, Stropharia rugoso-annulata Farlow, Hericium erinaceus, Agrocybe cylindracea with the use of molecular Markers are reviewed with emphasis on random amplified polymorphic DNA (RAPD), inter simple sequence repeat (ISSR), sequence-related amplified polymorphism (SRAP), sequence characterized amplified region (SCAR).The main results were as follows:1. We adopted the use of three PCR-based techniques, RAPD, ISSR and SRAP for 15 cultivated strains of Tricholoma giganteum massee. Clustering dendrogams were constructed using average linkage clustering. Based on cluster analysis, 15 cultivars were classified to 3 types at a distance of 12. Analysis of genetic distance showed that the genetic similarity coefficiency between cultivar 1 and 3 were high, they were all genetically related cultivars, and cultivar 12 is different from other cultivar. From the band patterns of the PCR products, clear and reproducible polymorphic fragments were selected 5 primers were designed and tested 5 SCAR primers were considered being converted into SCAR marker successfully. Based on cluster analysis of SCAR, ISSR, RAPD, SRAP .So it is proposed that 1 and 3 belong to the different names which are homogenous and cultivar 12 should not be Tricholoma giganteum massee.2. From the band patterns of the PCR products, clear and reproducible polymorphic fragments were selected. 18 primers were designed and tested. 17 scar primers were considered being converted into SCAR marker successfully. Based on cluster analysis of SCAR, ISSR, RAPD, SRAP. So it is proposed that 12 and 13 belong to the different names which are homogenous.4. We adopted the use of three PCR-based techniques, RAPD, SRAP and ISSR for 46 cultivated strains of Agrocybe cylindracea. Clustering dendrogams were constructed using average linkage clustering. Based on cluster analysis, 46 cultivars were classified to 7 types at a distance of 15.Analysis of genetic distance showed that the genetic similarity coefficiency between cultivar 32 and 37 were high, they were all genetically related cultivars. From the band patterns of the PCR products, clear and reproducible polymorphic fragments were selected.20 primers were designed and tested.20 scar primers were considered being converted into SCAR marker successfully. Based on cluster analysis of SCAR, ISSR, RAPD, SRAP. So it is proposed that 32 and 37 belong to the different names which are homogenous.
