Objective TO observe the inhibitory effect on growth of JianPiBuShen Decoction on micebearing Lewis lung cancer and investigate its mechanism.Methods Fifty-five C57BL/6J mice that had been successfully established Lewis lungcancer were randomly divided into five groups:model group,CTX group,The low dose ofJianPiBuShen group,CTX+The low dose of JianPiBuShen group,CTX+The high dose ofJianPiBuShen group. After14days, the inhibitory rate of tumor was calculated; the index ofspleen and thymus were calculated; the pathological changes of tumor tissue were observedby HE staining; the tumor tissue ultrastructure was observed by electron microscope; theexpression of VEGF,CD44V6and Ki-67were detected by immunohistochemistry.Results(1) Compared with the model group,the tumor weight of other groups reduce, withstatistical significance (P<0.01); The low dose of JianPiBuShen group,CTX+The low dose ofJianPiBuShen group,CTX+The high dose of JianPiBuShen group compared with CTX group,with statistical significance(P<0.05); CTX+The low dose of JianPiBuShen group comparedwith CTX+The high dose of JianPiBuShen group, with statistical significance(P<0.05).(2) Compared with the model group, the spleen and thymus index of The low dose ofJianPiBuShen group increase, the spleen and thymus index of CTX group reduce, Thedifference was significant (P<0.05); CTX+The low dose of JianPiBuShen group、CTX+Thehigh dose of JianPiBuShen group compared with CTX group, The difference was notsignificant (P>0.05).(3) Under the light microscope, The low dose of JianPiBuShen group、CTX+The low dose of JianPiBuShen group、CTX+The high dose of JianPiBuShen group compared with modelgroup, tumor cell form change, many cells were necrotic, the nuclear cracking; Under theelectron microscope, The low dose of JianPiBuShen group、 CTX+The low dose ofJianPiBuShen group、CTX+The high dose of JianPiBuShen group compared with modelgroup, tumor cell membrane structure was not complete, cell nuclear break, chromation setedge, visible apoptotic body.(4) The result show, compared with model group, The other groups can down-regulatedVEGF protein expression(P<0.05).(5) The result show, compared with model group, The other groups can down-regulatedCD44V6protein expression(P<0.05), CTX+The high dose of JianPiBuShen group wasoptimum.(6) The result show, compared with model group, The other groups can down-regulatedKi-67protein expression(P<0.05).Conclusions(1) JianPiBuShen Decoction can significantly inhibit the growth of Lewis lung tumor cellsin mice.(2) JianPiBuShen Decoction can protect immune organs, it can promote effect of the CTXand reduce toxicity of the CTX on disease.(3) JianPiBuShen Decoction Can induce cell apoptosis, which may be one of the anti-tumormechanisms.(4) JianPiBuShen Decoction Can down-regulated VEGF protein expression, inhibit theblood vessels formation of tumor tissue.(5) JianPiBuShen Decoction Can down-regulated CD44V6protein expression, inhibit theadhesion effect of tumor cells.(6) JianPiBuShen Decoction Can down-regulated Ki-67protein expression, inhibit theproliferation of tumor cells.
