Objective To explore the expression of FAK and its clinical significance in patients with acute leukemia (AL).Methods Isolating bone marrow mononuclear cells, and RNA was extracted by RNA centrifuge column method. The levels of leukemic cell FAK mRNA in50AL and10healthy controls were measured by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) technique. Chromosome specimens preparation of bone marrow cells were made using direct and short term culture, and karyo-typing was performed by R-banding technique.Results The expression positive rate of leukemic cell FAK mRNA in50patients with AL was66.0%, which was significantly higher than that in normal controls (20.0%)(x2=5.486, P<0.05). The expression positive rates and levels of FAK mRNA in ALL cells and ANLL cells were significantly higher than that in normal controls (X2=4.436, P<0.05; t=7.985、5.595, P<0.05), but the difference between ALL and ANLL was not significant(x2=0.0299, P>0.05; t=2.009, P>0.05). The expression positive rates and levels of FAK mRNA in AL patients at diagnosis and relapse were significantly higher than that in normal controls(x2=9.38, P=0.015; t=8.789, t=7.5756, P<0.05), but there was no statistical significance between them(P=1.000, t=1.010, P>0.05). The positive rates at remission was lower than that in initially-treated and relapse patients (x2=10.26, P=0.0167; t=4.4359, t=7.822, P<0.05), but there was no statistical significance as compared with normal controls(P=0.66, t=0.5879, P>0.05). The expression positive rate and level of FAK mRNA in the abnormal karyotype group were significantly higher than that in the abnormal karyotype undetected group (x2=5.82, t=6.72, P<0.05).Conclusions FAK mRNA was over expressed in leukemia patients, and may be related to curative effect of the disease. It may be used for leukemia treatment targeting the provision of new sites.
