ObjectiveThis paper determined the right medicinal part of Breynia fruticosa through the comparasion of the total flavonoids content in stem and branches of Breynia fruticosa, combined with the comparative study on the in vitro bacteriostasis in stem and tender branch-leaves water extraction liquid of Breynia fruticosa. On this basis, further to determine the antibacterial effective parts of the tender branches filtered water extraction liquid, and through the observation of the curative effect-chronic dermatitis eczema in mice, confirmed the therapeutic effect of aqueous extract of tender branch-leaves water extraction liquid of Breynia fruticosa, and screening the effective parts, for further to provide experimental basis study of the anti-inflammatory anti-itching effect mechanism and the development of new agents of Breynia fruticosa.Methods(1)Rutin was used as reference sample by UV spectrophotometry and5%NaN02-10%Al(N03)3-10%NaOH as color-developing agents and the measurement was conducted at511nm.(2)The content of Epicatechin in leaves of Breynia fruticosa was determined by using HPLC. Permeating time, ratio of solid to liquid, extraction time and extraction times for the affection of water extraction rate of Epicatechin in leaves of Breynia fruticosa were detected by the orthogonal test.(3)Bacteriostatic activities of the water and alcoholic extracts were tested by paper disc diffusion method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) to sensitive bacteria of the extracts of tender branch-leaves were evaluated by the liquid media two-fold dilution method. Using fluorescence methods, the water extracts of branch-leaves from Breynia fruticosa were divided into four parts, including chloroform, ethyl acetate, N-butyl alcohol and remaining water part, using liquid media two-fold dilution method, bacteriostasis experiment studies in vitro are maken respectively to staphylococcus aureus, resistant staphylococcus aureus and pseudomonas aeruginosa, of four different polar parts to screen effective parts of bacteriostasis, and determinate the sensitive bacteria MIC and MBC.(4) To observe the therapeutic effect on chronic dermatitis-eczema in mice for the water extracts of branch-leaves from Breynia fruticosa. Fifty cases of models with chronic dermatitis-eczema were induced with2,4-dinitrochlorobenzene on mice. Sixty mice were assigned into the substrate matched group, the model group, Piyanping group, the hign and low dose of the tested drug on average. All models were treated with corresponding drugs for12consecutive days, twice per day. Using vernier caliper measure and calculate the ear thickness difference; using electronic analytical balance to weigh and calculate the ear swelling degree and the inhibition rate; the anatomy of thymus and spleen to weigh and calculate the corresponding index; last the changes of ear histopathology in mice of each group were observed by optical microscope.Results(1)The total flavonoids in Stem and tinder branch-leaves of Breynia fruticosa were38.58mg·g-1and79.16mg·g-1.(2)The content of Epicatechin in leaves of Breynia fruticosa was determined by using HPLC. Permeating time, ratio of solid to liquid, extraction time and extraction times for the affection of water extraction rate of Epicatechin in leaves of Breynia fruticosa were detected by the orthogonal test. The content of Epicatechin was0.7031mg·g-1.(3) MIC and MBC of the water extracts of the branch-leaves to Staphylococcus aureus were15.625mg/ml and125mg/ml, and the antibacterial rate reached to257.0%; MIC and MBC of the water extracts of the branch-leaves to Escherichia coli were125mg/ml, and the antibacterial rate reached to115.5%; None of the extracts exhibited inhibitory activity against Candida albicans and also the water extracts of the stems to the tested bacteria. Ethyl acetate part showed the strongest bacteriostasis to three bacteria, all the MIC was62.5mg/ml, MBC was250mg/ml; Followed by were the N-butyl alcohol of water saturation part and remaining water part, the MIC of the tested strains were125mg/ml and62.5mg/ml, the MBC of N-butanol part were500mg/ml、500mg/ml and250mg/ml; Chloroform part had no bacteriostasis to the tested strains.(4)The tested drug for a certain dose can effectively relieve the ear thickening、swelling and significantly suppress the immune organ index, and improve its pathology change with a dose-dependent relationship. The tested drug has good treatment effect on chronic dermatitis-eczema in mice. The ethyl acetate high dose group and n-butyl alcohol group of water extracts of tender branches-leaves of Breynia fruticosa can obviously improve the model of chronic dermatitis, eczema of thickening of mouse ear, can significantly inhibit ear swelling in mice, inhibit the chronic dermatitis, eczema was detected in the lesions of mice inflammatory cell infiltration, improve the ear piece of pathological changes, n-butyl alcohol group significantly inhibit the immune organ index in mice. And chloroform part and the rest of the water area had no obvious effect of chronic dermatitis eczema model mice.ConclusionsThe above results showed that the medicinal part of Breynia fruticosa is its tender branches-leaves. The antibacterial effective parts and the effective parts of treatment-chronic dermatitis eczema in mice of tender branches water extraction of Breynia fruticosa were ethyl acetate and n-butyl alcohol parts. The research content in this paper could provide the basis for the anti-inflammatory and anti-itching pharmacological action mechanism and development of relevant preparations for Breynia fruticosa.
