| ObjectTo investigate the expression of Suppressor of cytokine signaling2(SOCS-2)and Signal transducers and activators of transcription5(STAT-5) in hepatocellularcarcinoma(HCC) and their relationship to clinicopathologial features.Methods84cases of fresh tissue of hepatocellular carcinoma had been collected fromJanuary2012to February2013at department of Hepatobiliary and PancreaticSurgery of Zhengzhou University. All cases are diagnosed by experiencedpathological doctor, and all of these cases have not been accepted any treatmentbefore operation. Some specimens are put into-80℃fridge to preserve for extractionof RNA for RT-PCR to detect the level of mRNA of SOCS-2and STAT-5in livercancer tissues and para-carcinoma tissues. Others are preserved in formalin andembedded in paraffin to make tissue slice to detect the level of protein of SOCS-2andSTAT-5in liver cancer tissues and para-carcinoma tissues by immunohistochemical.At last these data were analysised together with follow-up data and theclinicopathologic data Results1. The relative amount of SOCS-2mRNA in hepatocellular carcinoma andadjacent tissues, respectively, is0.089±0.034ã€0.340±0.083. The expression ofSOCS-2mRNA in carcinoma tissues is apparently lower than in adjacenttissues(P<0.05).2. The relative amount of STAT-5mRNA in hepatocellular carcinoma andadjacent tissues, respectively, is0.532±0.094ã€0.276±0.065. The expression of STAT-5mRNA in carcinoma tissues is apparently higher than in adjacent tissues(P<0.05).3. The results in the qualitative analysis to the protein of SOCS-2inhepatocellular carcinoma and adjacent tissues is that the expression of SOCS-2incarcinoma tissues is lower carcinoma tissues, while in adjacent tissues the result isadverse(P<0.05).4. The results in the qualitative analysis to the protein of STAT-5inhepatocellular carcinoma and adjacent tissues is that the expression of STAT-5incarcinoma tissues is higher carcinoma tissues, while in adjacent tissues the result isadverse(P<0.05).5. The relationship between the expression of SOCS-2with theclinicopathologial features of HCC: the expression of SOCS-2is low, while STAT-5is high in carcinoma. The expression is not significant correlation with the patient’sage, gender, the size of tumor, differentiated degree(P>0.05), and is significantcorrelation with the level of AFP, metastasis and portal venous cancerembolus(P<0.05).6. The relationship between the expression of STAT-5with the clinicopathologialfeatures of HCC: the expression of STAT-5is high in carcinoma. The expression isnot significant correlation with the patient’s age, gender, the size of tumor,differentiated degree(P>0.05), and is significant correlation with the level of AFP,metastasis and portal venous cancer embolus(P<0.05).7. The expression imbalance correlation of SOCS-2and STAT5: SOCS-2andSTAT-5expression was negatively correlated, the result of mRNA is r=-0.612,P=0.001(P<0.05), and the result of protein is r=-0.574,P=0.001(P<0.05). Conclusions1. STAT-5has some relation with the genesis and progress of HCC, STAT-5might be one oncogene of HCC.2. SOCS-2has some relition with the inhibition of the genesis and progess ofHCC, SOCS-2might be one cancer suppressor gene of HCC.3. The SOCS-2gene may have a negative regulation to the function of the geneSTAT-5in the progress of the genesis of HCC. |
