Objective:We put the human gastric cancer cells transplanted into nude mice to establish tumor Model, then the cell experiment and animal experiment will be carried out, to investigate the inhibitory effect of125I particles on gastric cancer.Tumor after125I radiation, we examined the apoptosis of cancer cells and angiogenesis in gastric cancer cases. To provide theoretical basis for125I particles implantation in treatment of malignant tumor.Methods:We put the human gastric cancer cells (BGC803) transplanted into nude mice to establish tumor Model,when the tumor grows to a diameter of about1.2cm. the nude mice were randomly divided into blank control group (n=30particles),control group (n=30) and experimental group (n=30only). Blank particle group was implanted with a blank particle (Omci). The experimental group were implanted with a125I particles of0.8mCi (lmCi=37MBq), the control group were not implanted particles. We put the particle implantation of tumor, then every three days to measure the size of the tumor, and to calculate the tumor volume, and then we use the tumor volume calculated the tumor inhibitory rate.25I particles were implanted in nude mice for twenty-eighth days when the mice were killed, we take tumor specimens. Weigh out the weight of tumor with electronic balance. Each specimen was divided into three parts.The sespecimens were treated with PBS preservation solution, liquid nitrogen, paraformaldehyde preservation. We used flow cytometry (Annexin VFITC/PI staining) technology to detect apoptosis.Then we used immunofluorescence to observe the tumor microvessel density. Finally, we use the Western blot and RT-PCR to detect the expression of tumor tissue VEGF. Results:1. Experimental group on the inhibitory rate of the control group was39%,experimental group on the inhibitory rate of the blank control group was38%.2. Flow cytometry results showed:compared with the control group, the experimental group has an obvious apoptosis.3. The results of immunohistochemistry detection showed:compared with the control group,, the vessel of the experimental group reduced. And the Expression of VEGF in vascular tissue cells decreased obviously.4.Dye fluorescence quantitative PCR detection results showed:compared with the control group, the expression level of mRNA VEGF gene in the experimental group was reduced, and there is significant difference.5. Results of Western blot technique showed that:compared with the control group,the expression level of VEGF gene in the experimental group was reduced,and there is significant difference.Conclusions:1.125I particles can significantly inhibit the growth of gastric cancer (BGC803).2.1251particles can not only kill cancer cells directly (BGC803), but also can promote the apoptosis of gastric cancer cells.3.125I particles can inhibit angiogenesis of gastric carcinoma.4.125I particles can significantly inhibit the expression of VEGF in gastric carcinoma.
