Senecio scandens Buch.-Ham.is a Chinese materia medica that has natural antiseptic antiphlogistic effect,it also has obvious inhibitory action on several types of bacteria such as taphylococcus aureus.Senecio scandens Buch.-Ham.was studied comprehensively in our research to reasonablely developt its medicinal value,quality control and regulate the market.Flavonoids, organic acid and volatile oil which are the main active ingredients of Senecio scandens Buch.-Ham.were extracted and separated to provide raw material for the development of new drugs.we processed the data combined with chemometrics.HPLC-DAD fingerprint and GC-MS fingerprint were established to reflect the chemical composition completely.The first fraction of this thesis was to extract and separate volatile oil in Senecio scandens Buch.-Ham.(chapter2).we had extracted volatile oil from Senecio scandens Buch.-Ham.by steam distillation extraction.And the different medicinal parts have been compared by using GC-MS and chemometrics.51compounds of volatile oil have been identified,these compounds accounteed for94.64%of the total.A reliable HPLC-DAD method was developed and validated for quantification of four ingredients(chlorogenic acid,rutin,hyperin and quercetin)inS.scandens Buch.-Ham from different parts and habitats.(chapter3). Extraction conditions and the chromatographic conditions for the determination of4ingredients have been established,the experiment adopt gradient elution,used HELP method in resolution of overlapping peaks,and quantifed with external standard method.This simple method had high precision and perfect reproducibility,and obtained satisfactory results in the determination of this herb.The third fraction of this thesis focused on non-volatile components analysis and volatile components analysis with high performance liquid chromatography-diode array detector (HPLC-DAD) fingerprint and GC-MS(gas chromatography)fingerprint of volatile components(chapter4). The content included optimization of extraction,selection of a suitable separation conditions for chromatographic,and validation of methodology,similarity analysis,PCA analysis and HCA analysis.The results showed that the fingerprints of constituents were very similar.The similarity was greater than0.95,but the main active component content has very big difference.Volatile components analysis and non-volatile components analysis results are basically identical,thus indicating origin have a great influence on the content of the main effective components. This experiment has established the HPLC-DAD fingerprint and GC-MS fingerprint, it is usefull for taking full control of the quality of traditional Chinese medicine.
