Objective:To analysise the relative expression and regularity of E-cadherin and Calmodulin mRNA in facial nucleus of Newzland rabbits after facial nerve crush injury.Then treating by electroacupuncture,to investigate the electro acupuncture therapy in the treatment of facial paralysis in the role of,and the expression of E-cadherin and Calmodulin in the facial nucleus,including the correlation between them,would be discussed.At last,illustrating the patterns and channels of electroacupuncture in the recovery process after facial nerve injury.Methods:Dividing 52 healthy adult New Zealand rabbits into normal control group,operation control group and acupuncture treatment group.The facial nucleus of 4 animals comprised the normal control group,without nerve injury and acupuncture treatment,would be cut off directly trial,while the right sidedness bucal branch of facial nerve crush injury(Sunderland IV)animal model to be established on acupuncture treatment group and operation control group(each 24 animals)on 1 day,4 day,7 day,14 day,21day and 28 day(each 4 individuals)after operation.At 2 hours after operation,acupoint electroacupuncture could went into effect in acupoint of Quanliao(颧缪)、Dicang(地仓)、Sibai(四白)、Yifeng(翳风)、Jiache(颊车)、Yangbai(阳白)and Hegu(合谷)on acupuncture treatment group,lasting 30 minute each time per day,until experimental time points.Meanwhile,there were none factor to inflect affect the recovery of operation control group.After cut off the pons and located the FM basis on the brain atlas of rabbit,HE staining was used to locate and observe the morphological changes of the facial motoneurons.Then approaching extracting total RNA,reverse transcription and PCR technology,detect the relative expression of E-cadherin and calmodulin in facial nucleus,and it would be statistical analysised adopt SPSS 17.0.Results:To compare with the relative expression quantity of E-cadherin and calmodulin on each time point between each group,the data of normal control group was the lowest one,and the acupuncture treatment groups’were higher than the operation control groups’(P<0.05).In both of acupuncture treatment group and operation control groups,the relative expression of E-cadherin mRNA have raised on the first day after operation,and there were maintenance period of high expression in each group during 7th~14th day.During this period,the relative expression have been increasing continuously until 14th day,and switching to steady decline.In operation control group,the relative expression of calmodulin m RNA has been increasing two weeks after operation,and turn to decrease after 14th day;but the early appearance of peak expression and extending maintenance period of high expression level should be showed in acupuncture treatment group.Applyed linear correlation to statistics the relative expression quantity of E-cadherin and calmodulin,there should be a positive correlation.Conclusions:After facial nerve injury,the expression of E-cadherin and calmodulin in facial nerve nucleus have both significantly increased,and probably can promote regeneration of neuron.One of ways that electroacupuncture affect on peripheral facial nerve injury possibly by increasing the expression of E-cadherin and calmodulin,advancing the expression peak,and prolonging the maintenance period of high expression.E-cadherin could promote calmodulin functions through cell signaling mediated by second messenger Ca2+possiblely.
