ObjectiveThe purpose of this study was to detect the expression of Pituitary tumor transforming gene(Pituitary tumor transforming gene,PTTG), Ki-67 nuclear antigen and Survivin protein in nasal sinuses adenoid cystic carcinoma and the normal nasal mucosa tissues by immunohistochemical method(SP method),and to analyze the correlation analysis of them,aiming to explore the molecular mechanism of PTTG and Ki-67 and Survivin in nasal sinuses adenoid cystic carcinoma to provide a scientific basis for the early diagnosis and prognosis of the disease. Materials and Methods1.Collection of 45 cases of surgical removal of the specimens of nasal sinuses adenoid cystic carcinoma in otolaryngology of the first affiliated hospital of zhengzhou university between May 30,2004-2014 as the experimental group,each specimen is under two pathology experts’ diagnosis,19 cases of male, 26 cases of female; 30-78 years old(average age 56 years old).All preoperative patients were with no history of long-term oral nonsteroidal anti-inflammatory drugs, had not been preoperative chemotherapy and biological target therapy, etc.Selection of 20 cases of nasal benign tumor resection in patients with nasal normal mucosa tissues as control group.2.Using the method of immunohistochemical method(SP) to test the expression and distribution of pituitary tumor transforming gene PTTG protein, Ki-67 nuclear antigen and Survivin protein in nasal sinuses adenoid cystic carcinoma and normal nasal mucosa tissues,to compare them expressed in the experimental group and control group in the positive correlation, and the relationship between the each two factors.And analysis the relationship of PTTG protein, Ki-67 nuclear antigen, Survivin protein and clinical pathological features of nasal sinuses adenoid cystic carcinoma.3.Statistical analysis, all the expermental results using SPSS17.0 software for statistical analysis.Count data by chi-square test, the relationship between PTTG and Ki- 67 and Survivin using Spearman rank correlation analysis.P < 0.05 was statistical significance. Results1.The expression of PTTG protein the positive expression of PTTG protein is mainly located in the nucleus or cytoplasm, in SNACC organization, PTTG protein positive expression rate was 71.11%(32/45), normal nasal sinus mucosa epithelial tissue of positive rate was 15%(3/20), the difference was statistically significant(χ2 = 17.56, P < 0.01). Histological grade III ~ IV level positive expression of PTTG is significantly higher than I ~ II level(P < 0.05); SNACC pathological classification in implementing structure expression is higher than in tubular structure, the structure of the cribriform(P < 0.05); Lymph node metastasis were significantly higher in without metastasis(P < 0.05); Positive expression of PTTG protein has nothing to do with age, gender, location(P>0.05).2.The expression of Ki- 67 nuclear antigen the positive expression of Ki-67 nuclear antigen in cancer cell nucleus, in SNACC organization, Ki-67 nuclear antigen positive rate was 75.56%(34/45), no positive expression in normal nasal sinus mucosa epithelial tissue, the positive expression between them was statistically significant difference(χ2 = 28.73, P < 0.01).Histological grade III ~ IV level of Ki- 67 nuclear antigen positive expression is significantly higher than I ~ II level(P < 0.05);SNACC pathological classification in implementing structure expression is higher than in tubular structure, the structure of the cribriform(P < 0.05);Ki- 67 nuclear antigen positive expression has nothing to do with age, sex, lymph node metastasis,location(P>0.05).The expression of Survivin the positive expression of Survivin protein mainly in the cytoplasm or cell membrane in cancer cells, in SNACC organization, Survivin protein positive expression rate was 66.67%(30/45), normal nasal sinus mucosa epithelial tissue of positive rate was 5%(1/20), the difference was statistically significant(χ2 = 21.11, P < 0.01).Histological grade III ~ IV level positive expression of PTTG is significantly higher than I ~ II level(P < 0.05);SNACC pathological classification in implementing structure expression is higher than in tubular structure, the structure of the cribriform(P < 0.05);Lymph node metastasis were significantly higher in without metastasis(P < 0.05);Positive expression of Survivin protein has nothing to do with age, gender, location(P>0.05).4.The relevance of the PTTG and Ki- 67 nuclear antigen and Survivin in nasal sinuses adenoid cystic carcinoma tissues expression Spearman’s correlation test, PTTG and Ki- 67 nuclear antigen expressed in SNACC organization positively correlated(P < 0.05);Ki- 67 nuclear antigen and Survivin expression in SNACC organization positively correlated(P < 0.05);PTTG and Survivin expression in SNACC organization positively correlated(P < 0.05). Conclusions1.Nasal sinuses adenoid cystic carcinoma incidence may be related to the abnormal increase expression of PTTG protein, Ki-67 nuclear antigen and Survivin. The expression of PTTG protein, Survivin in nasal sinuses adenoid cystic carcinoma is associated with lymph node metastasis, clinical stage and pathological type, Ki- 67 nuclear antigen is associated with clinical stage and pathological type.2.The expression of PTTG protein, Ki- 67 nuclear antigen and Survivin play synergy role in nasal sinuses adenoid cystic carcinoma, PTTG and Ki- 67 nuclear antigen protein, Ki- 67 nuclear antigen and Survivin and the expression of PTTG and Survivin protein in SNACC was significantly positively related to both.3.PTTG, Survivin protein and Ki- 67 play a very important role in the development of nasal sinuses adenoid cystic carcinoma, they can be used as a judgment and valuable molecular biological indicators of SNACC.The expression of them can be used to detect the malignant degree of SNACC and to evaluate the prognosis of patients.
