Expression And Significance Of SOX4 In Oral Lichen Planus Related Oral Squamous Cell Carcinoma

Objective:1.Use liquid chromatography-tandem mass spectrometry technic to screen out the high risk biomarker SOX4 which may cause oral lichen planus (OLP) malignant transformation.2.Detect the expression of SOX4 protein in oral squamous cell carcinoma(OSCC) cell line and normal human oral keratinocyte(NHOK) cell line at protein level.3.Detect the expression of SOX4 protein in OLP,OSCC and oral lichen planus related oral squamous cell carcinoma(OLP-OSCC) by immunohistochemistry.Methods:1.All of the formalin-fixed and parrffin-embedded(FFPE) tissue samples were provided by Sichuan Academy of Medical Sciences and Sichuan Provincial Peoples Hospital pathology department,wich include 27 cases of OSCC,19 cases of OLP and 5 cases of OLP-OSCC,all of them were the patients of stomatological departmet in 2004-2013.We screen out the differential protein SOX4 by LC-MS/MS.2.Use western blotting method to detected the expression of SOX4 protein in OSCC cell line and NHOK cell line.3.Detected the expression of SOX4 protein in 19 OLP,27 OSCC and 5 OLP-OSCC FFPE tissue sample by immunohistochemistry.Results:We identified 97 kinds of protein in OLP samples,135 kinds in OSCC samples and 142 kinds in OLP-OSCC,and screened out the SOX4 protein as the differential protein.2.We found SOX4 did not expressed in NHOK cell line,but we found strong expression of SOX4 in OSCC cell line.3.SOX4 protein was overexpressed in both OSCC and OLP-OSCC cytoblast,the positive rate were 96.29% and 100%,otherwise the positive rate of SOX4 protein in OLP smples was only 5.26%.Conclusion:1.We succeed in screenning out SOX4 protein as the critical differential protein from OLP.OSCC and OLP-OSCC FFPE tissue samples by use LC-MS/MS.2.SOX4 protein was found a highly expression in both OSCC tissue and OSCC cell line.3.We speculated that SOX4 may act as a new risk-mark,which play an important role in process of OLP turn into OSCC.
