The Study On The Effect Of Lipotoxicity In Myocardial Of Rats With High-fat Diet And The Intervention Effect Of Aerobic Exercise And Grape Seed Extract

Purpose:By comparing the change of FFA, IL-6,TNF-alpha level in plasma of rats and the level of TLR4, NF-κBm RNA, FFA, ICAM-1 in myocardial before and after experiment, explore the effect of lipotoxicity in myocardial of rats with high-fat diet and the protection of myocardial with aerobic exercise and its joint supplement of grape seed extract.Methods: Got 50 clean, male SD rats,8 week in age, were randomly divided into five groups after 1 week of acclimation: control group(N=10, group C), high-fat diet group(N=10, group H), high-fat diet exercise group(N=10, group HE), high-fat dietary supplement GSPE group(N=10, HG), high-fat dietary supplement GSPE and exercise group(N=10, HEG). Group HE and HEG with aerobic exercise. 0 slope are used treadmill exercise. Running speed:the 15m/min at the first week, 20m/min at the second week, after that, increase 1m/min a week. Times:50min/times. Sports Time: 21:00-24:00,1 time/day, 6 days/week, Sundays off, for six weeks. Group HG and HEG were administered GSPE(200mg/kg body weight), the other group fed the same amount of volume of saline water, before the half an hour of exercise to lavage. Record the weight and length every group for every week, in the end of the experiment, test the levels of FFA, IL-6, TNF- alpha in plasma and the levels of FFA, ICAM-1 in myocardial with ELISA, test the level of NF-kappa Bm RNA in myocardial with RT-PCR, test the level of TLR4 in myocardial with Western blot.Using SPSS17.0 software processing obtained data.Result: 1.Group H Lee’s index, FFA, TNF-alpha, IL-6 level in rats plasma and the level of TLR4, NF-κBm RNA, FFA, ICAM-1 in myocardial was significantly higher than group C(P<0.01).2.Compared with group H, the Lee’s index, FFA, TNF-alpha level in rats plasma ofgroup HE, HG, HEG was significantly decreased(P<0.01, P<0.05), no significant change of IL-6 in plasma(P>0.05), aerobic exercise and dietary supplement GSPE had no significant interaction(P>0.05).3.There is no significant change of ICAM-1 in myocardial between group H, HE,HG, and HEG(P>0.05), aerobic exercise and dietary supplement GSPE had no significant interaction(P>0.05).4.Compared with group H, the level of FFA, TLR4, NF-κBm RNA in myocardial was significantly decreased(P<0.05, P<0.01), There was a significant interaction between the two factors(P<0.05), meaning that at the same condition of GSPE supplement, aerobic exercise can make rats FFA significantly decreased(P<0.05), also,under the same movement condition, added GSPE can make the rats TLR4 and NF-кBm RNA level significantly decreased(P<0.01).Conclusions: 1.6 weeks high-fat diet can increase the weight of rats and Lee’s index in different degrees, accompanied by plasma FFA, TNF-alpha, IL-6 levels increased, the body showed lipotoxic and inflammation state.2.6 weeks high-fat diet can increase the the level of FFA, TLR4, ICAM-1 and NF-κBm RNA in myocardial, caused lipotoxic cardiomyopathy and increased arterial endothelial damage.3.6 weeks aerobic exercise and added GSPE can reduce rats obesity, lipotoxic cardiomyopathy and inflammation state, protect myocardial. Plus, combined with aerobic exercise and GSPE gives better results than simple aerobics or GSPE supplement.
