Objective: To explore the relationship between the proportion of myeloid-derived suppressor cells(MDSCs) and expression of Arg-1,i NOS,VEGF in serum of patients with peripheral T-cell lymphoma(PTCL),which would affect their prognosis prognosis.Methods : ⑴ The peripheral blood samples were taken from 25 patients with PTCL and 15 of healthy persons( the normal control group),and checked out the proportion of MDSCs/mononuclear cells in peripheral blood by flow cytometry, then analyzed the difference of proportion of MDSCs in patients with PTCL who had different clinical feature and those in healthy persons. ⑵ To analyze the expression level of Arg-1,i NOS,VEGF in serum of 25 patients with PTCL and 15 healthy people by ELISA.Results:⑴ There was a positive correlation between the proportion of MDSCs in peripheral blood and the expression level of Arg-1,i NOS,VEGF in serum in patients with PTCL.(2) ① The proportion of MDSCs in the peripheral blood of patients with PTCL wassignificantly higher than the healthy subjects [(4.59±1.41)%vs(1.36±0.59)%, P<0.05]. ② The proportion of MDSCs in peripheral blood of patients with PTCL was significantly lower postchemotherapy than pre-chemotherapy( 1.29± 2.95 %, P<0.05). ③ There was a significantly difference between the proportion of MDSCs and extranodal involvement, bone marrow involvement, stage, IPI score(P<0.05), and there was a significantly raise of proportion of MDSCs at the statuses of two or more extranodal involvement, positive bone marrow involvement,stage III/IV, middle or high risk of IPI score. There was no correlation between the proportion of MDSCs and age, LDH, volume of tumor(P>0.05).(3) ①The expression level of Arg-1, i NOS and VEGF in serum of patients with PTCL was significantly higher than the healthy subjects(17.09±2.56 ng/ml vs 10.93±1.08 ng/ml,P=0.000;21.90±3.65 Iu/ml vs16.39±6.08 Iu/ml,P=0.010;37.46±38.73 pg/ml vs 15.82±8.37 pg/ml,P=0.012). ②The expression level of Arg-1, i NOS and VEGF in serum of patients with PTCL was significantly lower postchemotherapy than pre-chemotherapy. ③ There was a significantly difference between the expression level of Arg-1, i NOS and VEGF in serum and extranodal involvement, bone marrow involvement, stage, IPI score(P<0.05), and there was a significantly raise of expression level of Arg-1, i NOS and VEGF at the statuses of two or more extranodal involvement, positive bone marrow involvement, large mass( ≥10cm), stage III/IV, middle orhigh risk of IPI score.Conclusion : ⑴ There was a positive correlation between the proportion of MDSCs in peripheral blood and the expression level of Arg-1,i NOS,VEGF in serum of patients with PTCL.(2) The proportion of MDSCs in peripheral blood and the expression level of Arg-1, i NOS and VEGF in patients with PTCL may be the marker of early diagnosis,prediction of chemotherapy effectiveness and prognosis。...
