Objective: To explore the role of protein expression of STAT3, β-catenin and VEGFin normal brain tissues and human glioma genesis with development, proliferation andinfiltration, analyze the relevance of the three with gliomaclinicopathological parameters(gender, age, glioma tumor location and WHO classification) as well as the correlation ofSTAT3, β-catenin and VEGF expressed in human glioma in the hope of guiding theevaluation of glioma malignancy and prognosis, providing a theoretical basis for the searchfor new targets for therapy of glioblastoma.Method: All of the106block cases were collected from Xuzhou Medical CollegeAffiliated First People’s Hospital of Lianyungang from January2009to December2012.Among of them, male cases number was62and female cases number44, and the ratio was1.4:1. The youngest patient was only11years old, the oldest78, and the average age46.4.All gliomas specimens were collected from neurosurgical resection and confirmed bypathological diagnosis. All of them were divided into four grades of Ⅰ, Ⅱ, Ⅲ, Ⅳ bydiagnosticians from Department of Pathology of the Lianyungang First People’s Hospitalbased on the latest WHO Classification of Tumours of the Central Nervous System(2007).Low-grade group(Ⅰ、Ⅱ)41cases and high-grade group(Ⅲ、Ⅳ)65cases, including13cases of level Ⅰ,28of levelⅡ,31of level Ⅲ and34of level Ⅳ. In contrast,10cases ofnormal brain specimens were obtained by intracranial decompression surgery. Theexpressions of STAT3, β-catenin and VEGF in human glioma tissues were detected byimmunohistochemical Envision method, statistical analysis were combined with clinicaland pathological data. SPSS19.0was used to process data with rank sum test and spearmancorrelation coefficients test according to date type, considering sense statistically(P<0.05).Results:1. STAT3positive expression in normal brain tissues was not found, but in glioma the rate was50.0%(53/106), considering that the difference with P <0.01makes sensestatistically. STAT3expression had relationship to WHO classification of glioma(P<0.05),and the ratios were30.77%(4/13),32.14%(9/28),61.29%(19/31),61.76%(21/34)respectively with levelsⅠ, Ⅱ, Ⅲ, Ⅳ. The expression of STAT3had no correlation togender, age and tumor location(P>0.05).2. β-catenin positive expression in normal brain tissues was not found, but in gliomathe rate was86.79%(92/106), considering that the difference with P <0.01makes sensestatistically. β-catenin expression had relationship to WHO classification of glioma(P<0.01), and the ratios were61.54%(8/13),82.14%(23/28),90.32%(28/31),97.06%(33/34)respectively with levelsⅠ, Ⅱ, Ⅲ, Ⅳ. The expression of β-catenin had no correlation togender, age and tumor location(P>0.05).3. VEGF positive expression in normal brain tissues was not found, but in glioma therate was58.49%(62/106), considering that the difference with P <0.01makes sensestatistically. VEGF expression had relationship to WHO classification of glioma(P<0.01),and the ratios were15.38%(2/13),39.29%(11/28),58.06%(18/31),91.18%(31/34)respectively with levelsⅠ, Ⅱ, Ⅲ, Ⅳ. The expression of VEGF had no correlation togender, age and tumor location(P>0.05).4. In glioma, positive expressions of both STAT3and β-catenin were53cases whilenegative expressions of the two were14, which showed positive correlation(r=0.801, P<0.01). Positive expressions of both STAT3and VEGF were50cases while negativeexpressions of the two were41, which showed positive correlation(r=0.844,P<0.01).Positive expressions of both β-catenin and VEGF were62cases while negative expressionsof the two were14, which showed positive correlation(r=0.864,P<0.01).Conclusion:1. The positive expression difference of STAT3in glioma and normal brain tissuemakes sense statistically. STAT3expression has relationship to WHO classification ofglioma, while has no correlation to gender, age and tumor location.2. The positive expression difference of β-catenin in glioma and normal brain tissuemakes sense statistically. β-catenin expression has relationship to WHO classification ofglioma, while has no correlation to gender, age and tumor location. 3. The positive expression difference of VEGF in glioma and normal brain tissuemakes sense statistically. VEGF expression has relationship to WHO classification ofglioma, while has no correlation to gender, age and tumor location.4. STAT3, β-catenin and VEGF expression in glioma were positively correlatedbetween any two of the three.
