Objective:To investigate the expressions of COX-2%MMP-9and VEGF in human glioma.Methods:The expressions of COX-2、MMP-9and VEGF in55cases with human glioma and8specimens with normal brain tissues were detected by immunohistochemical staining(S-P methods), studying by cytological observation.Results:(1)The Positive expression of COX-2wasn’t observed in the normal brain tissue and the rates of positive expression of COX-2in glioma Grade Ⅰ、Ⅱ group, and Grade Ⅲ、Ⅳ group were62.07%and96.15%respectively. The expression intensity in different glioma grade groups showed increasing trend by sequence, the difference was statistically significant (P<0.05).In further comparison of any two of them it can be found that the difference between any two groups was statistically significant (P<0.05).(2)The rates of positive expression of MMP-9in normal brain tissues, glioma Grade Ⅰ、Ⅱ group.and GradeⅢ、Ⅳgroup were0.00%,72.41%and92.31%respectively, the difference was statistically significant(P<0.05).In further comparison of any two of them, it can be found that the difference between any two groups was statistically significant(P<0.05)(3)The rates of Positive expression of VEGF in normal brain tissues, glioma Grade I.. II group, Grade Ⅲ、Ⅳ group were00.00%,79.31%, and96.15%respectively. With the intensity of expression increase by sequence, the difference was statistically significant(P<0.05).In further e comparison of any two of them, it can be found that the difference between any two groups was statistically significant(p<0.05).(4)Using Spearman rank correlation test, it can be confirmed that Pathologic classification was positively correlated with COX-2expression index(rs=0.734, P<0.05)and MMP-9expression index(rs=0.483), P<0.05) and negatively correlated with VEGF expression index(rs=0.518, P<0.05), these were all highly significant(P<0.05);COX-2expression was significantly positively correlated with MMP-9expression in glioma(rs=0.631.P<0.05).In same, COX-2and VEGF expressions were significantly correlated positively (rs=0.687, P<0.05).Conclusion:The expression intensity of COX-2, MMP-9and VEGF were increased with the glioma malignant grades, suggesting that they might be involved in the development and progression of glioma. There was definite correlation between the positive expression of COX-2、 MMP-9and VEGF in glioma, which indicated that the development of glioma was a result of the interplay of many factors. COX-2may induce the expression of MMP-9and VEGF to promote the formation of angiogenesis, the invasion and the metastasis of glioma.
