Objective:To explore the expression of DUSP6, CK19, Galectin-3 and HBME-1 in PTC,NGWPH and NG, and the value of the above-mentioned molecular markers single or combined detection in differential diagnosis of PTC. To find out one immune molecule or combined markers with best sensibility and specificity in the diagnosis of PTC.Methods::174 confirmed thyroid cases were collected from the Pathology Department of the People’s Hospital of Liaoning Province during 2010.8-2014.6, including 96 cases of PTC and 43 cases of NGWPH and 35 cases of NG. Immunohistochemical staining combined with the technology of tissue chip were performed in the selected specimens to examine the expression of DUSP6, CK19, Galectin-3 and HBME-1. The formulas were used to calculate the sensitivity, specificity, positive predictive value, negative predictive value and diagnosis accurate rate of each immune molecular markers and the marker combinations. All the results were analyzed by SPSS statistical software. The level of significant difference was P<0.05.Results:1. The positive expression rates of DUSP6, CK19, Galectin-3, HBME-1 in 96 cases of PTC were 87.5%, 100%, 96.9%, 98.9%, and the rates in 43 cases of NGWPH were 11.6%, 39.5%, 20.9%, 11.6%, respectively. While compared the two groups, each molecular marker expression differences were statistically significant(all P<0.01).2. The sensitivity of DUSP6, CK19, Galectin-3 and HBME-1 on the diagnosis of PTC were 87.5%, 100%, 96.9%, 98.9%, and the specificity were 88.4%, 60.5%, 79.1%, 88.4%, respectively. Compared of the four markers, the area under ROC curve of HBME-1 was the highest(AUROC=0.937;95%CI,0.880-0.994),and the second was Galectin-3(AUROC=0.880;95%CI,0.806-0.954) and DUSP6(AUROC=0.879;95%CI,0.811-0.947).What’s more, when the markers were combined examined, we found that it’s the best to use HBME-1 combined with DUSP6 and Galectin-3 triple positive expression as PTC diagnosis standard, since the sensitivity was 100% and the specificity was 89.5%(AUROC=0.947;95%CI,0.896-1.000).3. The expression of DUSP6 in PTC had significant correlation with the tumor size and lymph node metastasis(all P<0.05).However, there was no correlation between the expression of CK19,Galectin-3,HBME-1 in PTC and the clinical pathological characteristics(all P>0.05).Conclusion:1. DUSP6 can be used as an important immune molecule marker to predict the high risk of PTC.2. There was certain diagnostic value of DUSP6, CK19, Galectin-3 and HBME-1 on the diagnosis of PTC, among which HBME-1 had high sensitivity and specificity and it would be better to combined with DUSP6 and Galectin-3.
