Objective: This experiment intends to establish diabetes eye animal models by injection of Streptozotocin(STZ) to abdominal cavity, to explore the Osteopontin(OPN) and integrin αvβ3 expression level in the retina of diabetic rats. Preliminary discuss the mechanism of osteopontin and integrin αvβ3 on diabete retinopathy progress.Methods: Make similar mankind I type diabetes rat animal model. Sixty healthy and clean SD male rats were selected in this study, weighing 200g±10g, excluded other eye diseases,ordinary feeded, 15 rats were chosen for the blank control group randomly(group I), which were injected buffer solution intrapeitoneally, ordinary feeded for 20 weeks. The remaining three groups were proposed diabetes rats model group, fifteen rats each group and a total of forty-five, fasting for 12 hours, intraperitoneal injection of streptozotocin(STZ) at a dose of 55mg/kg to product diabetic rats model, blood glucose were measured after 72 h then. And the DM model was identified as the blood glucose ≥16.5 mmol/L after intraperitoneal injection of STZ. DM rats model also represent polyuria and polydypsia. All groups were ordinary feeded after injection. Respectively feeding group II,III and IV 12 weeks, 16 weeks, 20 weeks, after the experiment, all rats were sacrificed, retinal tissues were obtained and were given HE, immunohistochemistry stain, to detect the expression of OPN and integrin αvβ3 in retina respectively.Results:(1)Changes of body weight and blood glucose: Body weight in the normal control group(group I) were significantly higher than diabetic group(group II,III and IV)(p<0.05); but there were no significant difference between group II, III and IV. Compared with group I, plasma glucose was significantly higher than that in group II, III and IV(P<0.05), but there were no significant difference between group II, III and IV.(2) Immunohistochemistry staining: OPN and integrin αvβ3 positive cells performance for cytoplasm shader is tan-yellow, and expressed mainly in the ganglion cells layer cytoplasm and endothelial cells, the development as the illness in diabetic groups, the retina kernel layer cells have positive expression. Group II, III and IV of positive expression were higher than the group I(P<0.05): and there were also significant difference between group II, Ill and IV(P<0.05).(3) HE staining: Compared with the normal control group(group I), the number of breakthrough internal limiting membrane cells of DM rats model group(group II, III, IV)was significantly increased, and have statistical significance(P<0.05).Conclusion:(1)Diabetes rats’ retina have already appeared an obvious change for 12 weeks: the retina thickness become thin, didn’t see juxtavascular cell.(2)OPN and integrin αvβ3 express positive in diabetes retina,and increasing with the development progress of diabetes, which infects the diabetes retina may be related to the increased expression of OPN and integrin αvβ3.(3)OPN may have an interaction with αvβ3. It may play an important role in upgrading the expression of αvβ3 in DR.
