Objective:To study the different doses of the extractive of Bupleurum and Airpotato yam root in rat about liver toxicity related indexes, liver microsomal CYP450 enzyme activity, liver microsomal CYP450 enzyme protein expression, and discuss the mechanisms of hepatotoxical injury in rat.Methods:Take the different doses of the extractive of two kinds traditional Chinese to rat, and determine blood biochemical index (ALT. AST), oxidation index (T-AOC、SOD、MDA、GST)、 mitochondrial index (GLDH). cholestasis related index (TBA) and pathological changes of index; Establish in vitro liver microsomal drug metabolism system, using HPLC determine the decrease of the probe medicine after hatch each rat liver microsome to evaluate liver microsomal CYP450 enzyme activity; Apply Western blot to detect the protein expression of the rat liver microsomal CYP1A2、CYP3A1. CYP2E1.Results:The extractive of Bupleurum root to rat by light microscopy found the degree of liver disease in medium and large dose group is increased significantly to the blank group(p<0.01), and the degree of liver disease in small dose group is increased to the blank group(p< 0.05); Electron microscopy found that of each dose group compared with blank of mitochondria, the endoplasmic reticulum damage more serious, cytoplasmic lipid drops significantly increase; Blood biochemical index (ALT. AST), oxidation index (T-AOC. SOD. MDA. GST), mitochondrial index (GLDH), cholestasis related index (TBA) in each dose group compared with blank are found no statistical difference; Each dose group of CYP1A2. CYP2E1. CYP3A1 enzyme activity compared with the blank group are found no statistical differences (P>0.05); Each dose group of CYP1A2. CYP2E1. CYP3A1 protein expression compared with the blank group are found no statistical differences (P>0.05).The extractive of Airpotato yam root to rat by light microscopy found the degree of liver disease in large dose group is increased to the blank group(p< 0.05); Electron microscopy found that of each dose group compared with blank of mitochondria, the endoplasmic reticulum damage more serious, cytoplasmic lipid drops significantly increase; Blood biochemical index (ALT. AST), oxidation index (T-AOC、SOD、MDA、GST), mitochondrial index (GLDH), cholestasis related index (TBA) in each dose group compared with blank are found no statistical difference;Compared with that in the blank group,CYP2E1 enzyme activity in large dose group are obviously lower,but the small and medium dose group compared with blank are found no statistical difference;Compared with that in the blank group,CYP1A2、CYP3A1 enzyme activity in each dose group are found no statistical difference (P>0.05); Compared with that in the blank group, CYP1A2、CYP2E1、CYP3A1 protein expression in each dose group are found no statistical difference (P>0.05).Conclusion:Compared with that in the blank group, pathology in each dose the extractive of Bupleurum root group are obviously serious,but other each indexes are found no statistical difference,we reasoned that each dose of the extractive Bupleurum root to rat didn’t cause serious damage. So,the liver toxicity of the extractive of Bupleurum root remains further study.Compared with that in the blank group, pathology in large dose of the extractive of Airpotato yam root are obviously serious,but other each biochemical indexes are found no statistical difference,the large dose group can inhibit liver microsomal CYP2E1 enzyme activity,so the extractive of Airpotato yam root produce liver damage reason is inhibiting liver microsomal CYP2E1 enzyme activity, lead to drugs and their metabolites in the body for a longer time, resulting in rat liver damage.
