Effects Of Chinese Angelica Root, Astragalus Root, Bupleurum Root And Their Combination On Expression Of LN, PCⅢ, α-SMA And TGF-β1 On Experimental Hepatic Fibrosis In Rats

Objective:This experiment will evaluate the effects of Astragalus Root,Chinese Angelica Root,Bupleurum Root and their combination on experimental hepatic fibrosis (hepatic fibrosis index:LN,PCⅢ,α-SMA,TGF-β1)in Wistar rats.The exact effects of the interaction exists and the dose-reaction relation of the three traditional Chinese herbs are applied to treat the CCl4-induced hepatic fibrosis of rats will be concluded.The results will offer experimental reference to the clinical application.Methods:The experiment is composed of two parts(including,Part A and Part B). we choose 72 healthy male rats to divide into 18 groups by randomization, 4 rats in each group(including,each 8 intervention groups in Part A and Part B,a normal group and a model group).Hepatic fibrosis model is induced by injctionn of CCL4,hypodermic injectoin,one times every four days,42 days,Rats in intervention group are given drugs with different dosages at first injection of CCL4. A experimental design adopts factorial design.The two levels of part A are having and not having drugs.B experimental design adopts orthogonal design.The two levels of part B are high and low dosages. At the end of treatment ,all of the rats would be killed to test hepatic fibrosis index. the liver tissue is taken to make the HE staining ,VG staining and take photos.Results:①LN:The elevated serum LN in Chinese Angelica Root and Astragalus Root were markedly decreased.The elevated serum LN in Bupleurum Root was not significantly decreased.The combination of Chinese Angelica Root and Astragalus Root had interaction.The decreased effect of low dosage of Chinese Angelica Root was more obvious in comparison with high dosage of it.The decreased effect of high dosage of Astragalus Root was more obvious.②PCⅢ:The elevated serum PCⅢin three herbs alone was markedly decreased.The combination of three herbs had certain interactions.The decreased effect of high dose of Chinese Angelica Root and Astragalus Root was more obvious in comparison with low dose of it.The decreased effect of low dose of Bupleurum Root was more obvious.③α-SMA:The elevated serumα-SMA in three herbs alone was markedly decreased.The combination of Chinese Angelica Root and Astragalus Root had interaction.The decreased effect of low dosage of Chinese Angelica Root and Bupleurum Root was more obvious in comparison with high dosage of it.The decreased effect of high dosage of Astragalus Root was more obvious.④TGF-β1:The elevated serum TGF-β1 in three herbs alone was markedly decreased.The combination of...
