ObjectiveObserve the effect of the cluster needling of scalp acupuncture and the cluster needling of scalp acupuncture pretreatment on neurological function and expression of MAG protein in different time points in rats with acute focal cerebral ischemia.Explore the possible mechanism of scalp acupuncture treatment for ischemic stroke from the perspective of behavioristics and Proteomics.Methods72 healthy clean-grade male SD rats were randomly divided into three groups,named the false operation group(A,n=18),model group(B,n=18),the cluster needling of scalp acupuncture group(C,n=18),and the cluster needling of scalp acupuncture pretreatment group(C,n=18).Every group was divided into three groups(6 rats in each group)according to time points of 24h,3d,7d.The local cerebral ischemia rats model in Group B,Group C and Group D were prepared by modified suture-occluded method,while Group A was sham model.Rats in Group C were treated by the cluster needling of scalp acupuncture after the modeling,rats in Group D were treated by the cluster needling of scalp acupuncture 7 days in advance before the modeling,by contrast,Group A and B were intervened without acupuncture treatment.The rats in each group were evaluated by Zea-Longa's scoring system after being operated and being killed.Detect and identify the expression of MAG protein in the brain hippocampus tissues of MCAO rats in each time point by western blot methods.Observe the effect of the cluster needling of scalp acupuncture and the cluster needling of scalp acupuncture pretreatment on the expression of MAG protein,and explore the possible mechanism of scalp acupuncture treatment and scalp acupuncture treatment pretreatment for ischemic stroke.Results1.Neurological scoreIntra-group comparison:The first time Zea-Longa neurological score was performed 4 hours after surgery for each group of rats:compared with group A,the neural function scores of the group B,group C were significantly increased,and the difference was statistically significant(P<0.01).Compared with the group B,there was no significant difference in the neurological function score of group C(P>0.05);the neurological function score of group D was significantly reduced,and the difference was statistically significant(P<0.01).Compared with group C,the neurological score of group D was significantly reduced,and the difference was statistically significant(P<0.01).The second time Zea-Longa neurological score was performed for each group of rats in 24h,3d,and 7d,just before their sampling:(1)In 24h,3d,and 7d point:compared with group A,the neural function scores of group B,group C and group D were significantly increased,and the difference was statistically significant(P<0.01).(2)In 24h point:compared with group B,there was no significant difference in neurological scores in group C;The neural function score of group D was decreased,and the difference was statistically significant(P<0.05).Compared with group C,there was no significant difference in the neurological scores of group D(P>0.05).(3)In 3d point:compared with group B,there was no statistically significant difference in neurological scores between the group C and group D(P>0.05).Compared with group C,there was no significant difference in the neurological score of group D(P>0.05).(4)In 7d point:compared with group B,the neurological score of group C was significantly reduced,and the difference was statistically significant(P<0.01);there was no significant difference in the neurological scores of group D(P>0.05).Compared with group C,there was no significant difference in the neurological scores of group D(P>0.05).Inter-group comparison:In group C:compared with the 24h subgroup,there was no significant difference in neurological scores between the 3d and 7d subgroups(P>0.05).In group D:compared with the 24h subgroup,there was no significant difference in neurological scores between the 3d and 7d subgroups(P>0.05).2.Expression of protein MAGIntra-group comparison:(1)In 24h point:compared with group A,the expression level of MAG protein in group B was increased,and the difference was statistically significant(P<0.05);the expression level of MAG protein in group C was increased significantly,and the difference was statistically significant(P<0.01);there was no significant difference in the expression level of MAG protein in group D(P>0.05).Compared with group B,the MAG protein expression level in group C was not significantly different(P>0.05);the MAG protein expression level in group D was reduced,and the difference was statistically significant(P<0.05).Compared with group C,the MAG protein expression level in the scalp group D decreased significantly,and the difference was statistically significant(P<0.01).(2)In 3d point:compared with group A,the expression level of MAG protein in group B was increased,and the difference was statistically significant(P<0.05);group C and group D showed no significant difference in MAG protein expression level(P>0.05).Compared with group B,there was no significant difference in the expression of MAG protein between group C and group D(P>0.05).Compared with group C,there was no significant difference in the expression of MAG protein in group D(P>0.05).(3)In 7d point:compared with group A,the expression level of MAG protein in group B was increased,the difference was statistically significant(P<0.05);group C and group D showed no significant difference in MAG protein expression level(P>0.05).Compared with group B,the expression of MAG protein in group C was decreased,and the difference was statistically significant(P<0.05);the MAG protein expression level in group D showed no significant difference(P>0.05).Compared with group C,there was no significant difference in the expression of MAG protein in group D(P>0.05).Inter-group comparison:Compared with 24h point,the expression of MAG protein in 3d point showed no significant difference(P>0.05).Compared with 24h point,the MAG protein expression level of 7d point was reduced and the difference was statistically significant(P<0.05);there was no significant difference in the expression level of MAG protein between 7d point and 3d point(P>0.05).The expression levels of MAG protein in the other groups were not significantly different in each time window(P>0.05).Conclusion1.The method of cluster needling of scalp acupuncture can significantly reduce the neurological score of MCAO rats,improve the motor dysfunction of rats,and has a significant promotion effect on the neurological recovery in rats with cerebral ischemia.2.The method of cluster needling of scalp acupuncture pretreatment can inhibit the increase of the neurological score of MCAO rats in the acute phase after cerebral ischemia,reduce the neurological damage of cerebral ischemic rats,and show preventive and protective effects in cerebral ischemic rats.3.The expression of MAG protein increased significantly after acute cerebral ischemia,and the expression of MAG protein was significantly decreased after the intervention with cluster needling of scalp acupuncture.It showed that cluster needling of scalp acupuncture can improve the recovery of neural function after cerebral ischemia was promoted and brain protection was exerted.4.The expression level of MAG protein of MCAO rats intervened by cluster needling of scalp acupuncture pretreatment were significantly reduced in early stage after cerebral ischemia.It suggested that cluster needling of scalp acupuncture pretreatment can improve the tolerance of cerebral ischemia and inhibit the further damage of nerve function.
