Objectives By investigating the conditions of anxiety-depression in functional dyspepsia group and the normal control group, measuring the expression of Gastric and Somatostatin in antrum mucosa immunhistochemical in the two groups, then exploring the conditons of the psychosocial factors as well as the expressions of GAS and SS in two groups, thus we can get the impact of psychosocial factors on the expression of GAS and SS in antrum mucosa.Providing valuable clues for the treatment of FD.Methods 1 We chose 30 numbers of FD patients as the case group who visited the North China University of Science and Technology gastroenterology clinic and inpatient department from October 2014 to June 2015.All patients conformed to the RomeⅢdiagnostic criteria.15 were Epigastric Pain Syndrome patients, while the other 15 patients were Post-prandial Distress Syndrome patients. In the same period, 30 healthy people with no digestive tract symptoms were selected as control group..2 The basic data of the research objects were statistically analyzed using the general situation questionnaire(self made) and making the assessment of objects psychological state by the SAS and SDS.3 Before gastroscopy, all the subjects had to sign the "informed consent of the digestive endoscopy". When the examination is completed, then used the disposable biopsy forceps in the gastric antrum to take 1 pieces of tissue as a sample. Quickly fixed the samples in 10% formalin and embedded and made them sliced,.the general situation of gastric antrum mucosa was observed by HE staining, to observe the expression of gastric antrum mucosa GAS and SS by immunohistochemical staining(SP method). The positive expression of GAS, SS in antrum mucosa were observed by immunohistochemical methods, and the strength of the expressions were measured by density mean through the IPP image analysing software.4 Finally, we used SPSS 20 software to analyze all the data statistically. Before the the data is carried out, all the data were tested for normal distribution and homogeneity test of variance. Measurement data was expressed by mean standard±deviation. Using t test for comparison between two sample means; Using chisquare test for count data.We considered P﹤0.05 statistically significant, and P﹤0.01 statistically significant evidently. Results 1 The scores of SAS(52.03±5.67vs38.60±6.56)and SDS(51.70±6.87vs36.27± 6.27) in the FD group were both higher than those in the control group, the differences were statistically significant evidently(P﹤0.01). But there was no significant difference between the two subtypes of SAS(50.20±4.91vs53.87±5.94) and SDS score(50.33±7.48vs53.07±6.15)(P﹥0.05).2 The density mean of GAS in the antrum mucosa of FD group were lower than that in the control group(49.16±11.69vs59.78±14.98),the difference were statistically significant(P﹤0.01); There was no significant difference in expression of GAS between the two subtypes of gastric antrum(P﹥0.05). While the density mean of SS in the antrum mucosa of FD group were higher than that in the control group(9.35±2.30vs6.24±1.81), the difference was statistically significant evidently(P﹤0.01); There was no significant difference in expression of SS between the two subtypes of gastric antrum(P﹥0.05).3 What was more, we found that GAS immunohistochemical expression in antrum mucosa of patients with anxiety and depression was lower than those without anxiety and depression in the case group(44.41±8.46vs60.24±10.92),the difference was statistically significant evidently(P﹤ 0.01); However,SS immunohistochemical expression had the oppisite result(10.17±2.10vs7.43±1.52),and difference was also statistically significant evidently(P﹤0.01).Conclusions 1 FD patients are likely to have anxiety-depression state. 2 The expression of GAS is low in antrum mucosa of FD patients, however the expression of SS is high, and there is unbalance expression of brain-gut peptide in FD patients. 3 The imbalance of GAS and SS expression in gastric antrum is more severe in FD patients with anxiety and depression, suggesting that the imbalance of GAS and SS expression in gastric antrum mucosa of FD patients was related to mental and psychological factors.
