The Chemical Constituents And Anti-Scald Activity Of Leaf Of Cherokee Rose

Leaf of Cherokee Rose. is the leaf of Rosa laevigata Michx. in Rosaceae. In the "East Fujian Materia Medica", the dried powder leaf of Cherokee Rose. was recorded to cure burn and scald when used together with benne oil.So as a herb for treatment of traumatic hemorrhage and burn, it was widely used. Here we separeted nine compounds from Leaf of Cherokee Rose and established the structure elucidation about four compounds named. and made the methodology of content determination about the compoud of catechin, and did the pharmacology activity screening of Leaf of Cherokee Rose about resistance to scald.We established a RP-HPLC method for the determination of catechin in the leaf of Rosa laevigata Michx. The GraceSmart RP18 column was used. The mobile phase was consisted of water(1% acetic acid was contained)-methanol (90:10). The flow rate was1.0 mL/min and the detection wavelength was 279 nm. The Results are:The linearrange is 0.1 mg/mL-1.0 mg/mL, r=0.9999. The average recovery and the RSD are 100.41% and 1.13% respectively. So the method appeared to be simple,accurate and reliable. It can be used as a quantitative method for Leaf of Cherokee Rose quality control.According to Experiment Mcthodology on Pharmacology of TCM and Methodology on Modern Times Phartnacology of ExPeriment, we designed three experiments in odrer to study the therapeutic effect of Leaf of Cherokee Rose:(1) Stimulation experiment on rats’skin to observe the stimulation of Leaf of Cherokee Rose. (2) Experiment of mice ears conrer induced inflammatory by Dimethyl Benzene to observe the efects of Leaf of Cherokee Rose in anti-inflammation. (3) Make the model of second degree scald, observe the effects of Leaf of Cherokee Rose on the scald wound rats. And The follow results were obtained:(1) Leaf of Cherokee Rose’s different extracts had not any stimulation to nomral rats’skin or cankerous skin. (2) Leaf of Cherokee Rose’s different extracts had obvious anti-inflamatory effect in the experimental of mice ears cornor induced inflamatory by Dimethyl Benzene, and the part using water extracted was the best. (3) Leaf of Cherokee Rose’s different extracts had the powerful effects of promoting scald wound healing, and the part using water extracted was the best. Through the observation on the skin irritation, anti-inflammatory activities and the effect to the model of second degree scald, we preliminary know that Leaf of Cherokee Rose’s different extracts have different degrees of activity to the scald rat models.And the part using water extracted was the best.All in all, we can obtained Leaf of Cherokee Rose has the efects of anti-inflammatory and promoting scald wound healing.It offers foundation for clinic curative effect and display the drug excellent prospect on application.
