Objective To investigate the effects of corn peptide and aerobics on the levels of metablic hormones such as insulin, leptin, glucagon, MCP-1, PP and PYY in plasma of obese rats in order to analyze the hormone related mechanism of the reduction of body weight in rats.Methods 150 male SD rats were randomly divided into a high-fat feed group(H, n=135) and an ordinary feed group(C, n=15). The two groups were given high fat diet and normal diet respectively. 8 weeks later, the 48 obese rats were randomly divided into obesity susceptibility group(OS,n=8), obesity control group(OC, n=8), casein supplement group(Cas,n=8), corn peptide supplyment group(CP, n=8), obesity exercise group(OE, n=8) and corn peptide combined with aerobic exercise group(OEC, n=8). 8 rats of the obesity resistant ones were randomly picked out as the obesity resistant group(OR). In the next 4 weeks, we gave the CP and OEC group rats corn peptide diet, and gave the Cas group rats casein diet and gave the other groups ordinary diet. OE and OEC group rats took aerobic exercise(treadmill running) for 4 weeks. The levels of leptin, insulin, glucagon, MCP-1, PP and PYY in plasma were determined by a multiplex rat metablic magnetic bead array system according to the manufacturer’s instructions.Results 1) The effects of the 8 weeks high-fat diet on the obese rats(1) Before the high-fat diet intervene, there was no difference among all the rats. 8 weeks later, the weight, the visceral fat coefficient and the Lee’s index of OS group were both significantly higher than C group and OR group.(2) The levels of leptin, insulin, glucagon and MCP-1 in plasma of OS group rats were significantly higher than C group and the levels of leptin, insulin were significantly higher than OR group, which suggested that obesity can lead to enhance the levels of leptin, insulin, glucagon and MCP-1 in plasma, but high-fat diet couldn’t enhance the levels of leptin, insulin and MCP-1 in plasma of the obesity resistant ones.There were no differences among OS group, OR group and C group. But the levels of PYY in plasma of OS group and OR group rats were lower than C group,which suggested that hagh-fat diet might improve appetite by inhibitoring the release of PYY.2) The effects of the 4 weeks interventions on the obese rats(1) During the 4 weeks treadmill running, the weights of OC group, Cas group, CP group and C group were all improving, but the weight of OEC group was reducing.4 weeks later, the weight of OC group was significantly higher than C group, OE group and OEC group. The Lee’s index and the visceral fat coefficient of OE group and OEC group were both significantly lower than OC group, while the OEC group was the lowest. These results suggested that corn peptide combined with aerobic exercise might more conducive to losing weight.(2) After 4 weeks treadmill running, the levels of leptin and insulin in plasma of OEC group rats were significantly lower than OC group rats. Compared with OC group, the level of glucagon in CP group rats and the levels of MCP-1 in CP, OE, OEC and C groups rats were also significantly reduced. The PYY level of OC group rats were significantly lower than C group rats. To the contrary, the PYY level of OEC was significantly higher than OC and CP groups. The PP levels had no significant difference among all groups. These suggested that aerobic exercise combined with or without corn peptide could both reduce the levels of insulin, leptin and MCP-1 in the plasma of obese ones. In addition, corn peptide combined with aerobic exercise might reduce the intake by enhance the level of PYY.Conclusion(1) 8 weeks high-fat diet can induce 35.6% SD rats get fat.(2) Aerobic exercise combined with or without corn peptide can both loss weight and fat and aerobic exercise combined with corn peptide is more effective.(3) High-fat diet can improve the levels of leptin, insulin, glucagon and MCP-1 of obesity susceptibility ones and also improve the level of glucagon and reduce the level of PYY of the obesity resistant ones. These suggest that the obesity induced.(4) Corn peptide can reduce the level of glucagon and MCP-1. If corn peptide combined with aerobic exercise,it will be more effective. Corn peptide combined with aerobic exercise can improve the level of PYY, which may be the mechanism of energy expenditure and lipid oxidation leading to weight lose.
