Objective:In this study using an experimental of obstructive jaundice animal model, we are mechanism of NF-κB signaling pathway in mice with obstructive jaundice induced liver injury and elaborate joint motion Polysaccharides in the effect of improving liver injury,in order to provide a new basis for the feasibility experiments, and theoretical direction for the adjuvant treatment of obstructive jaundice.Methods:50 KM mice were randomly divided into five groups: sham surgery group 10(group B), model group 10(M group), medlar Group 10(G group), aerobic exercise group, 10(Y group), wolfberry-exercise group 10(G + Y group). In addition to the sham group which are in the common bile duct to the abdominal wall hanging 48 hours to build mouse obstructive jaundice induced liver injury model. Wolfberry group, wolfberry movement in the common bile duct ligation group were two days black fruit wolfberry polysaccharide orally fixed time orally once dose 0.3 ml / second, a total of six weeks. Aerobic exercise group, Chinese wolfberry-exercise group mice slope 0%, velocity 10 m / min, every day 30 min, six days a weekNo weight training treadmill exercise program, a total of six weeks of moderate intensity treadmill training. After the intervention of mice were fasted overnight, before the drawn first weighed, after anesthesia completely isolated liver tissue,, application hematoxylin- eosin(HE staining)Observation of pathological changes under microscopic structure of each group, mouse liver tissue; immunohistochemical method to detect liver NF-κB P65 protein, TNF-α in; Realtime-PCR technology to detect NF-κB, TNF-α, IKK-α,IkB-α expression. biochemical analysis serum total bilirubin(TBIL), alanine aminotransferase(ALT) and aspartate aminotransferase(ALT) content;Results: 1.serological results:Wolfberry+exercise group significantly reduced TBIL, ALT, AST levels compared with the model group(P <0.05), wolfberry group, the aerobic exercise group TBIL, ALT, AST levels were also significantly decreased(P <0.05).2. HE staining showed: Sham group of mice liver cell cords arranged radially; model group mice of liver cells is atrophy, hepatic cord rupture, nuclear condensation, etc., interstitial visible red blood cell accumulation; aerobic exercise group and wolfberry group were observed in liver irregular lobular architecture, hepatic cord neatly arranged owe, local shows steatosis cells; Wolfberry + exercise group was most significant, lobular complete, no distortion and necrosis.3.Immunohistochemical staining showed:Compared with the sham surgery group, the positive expression of NF-κB P65 and TNF-α were increased significantly in the model group(P<0.05).Compared with the model group,the positive expression of NF-κB P65 and TNF-αwere decreased evidently in the wolfberry + exercise group(P<0.01).4.Real-time PCR results showed that:Compared with the sham surgery group, the positive expression of NF-κB,TNF-α, IKK-αmRNA was decreased significantly in the model group(P<0.05).Compared with the model group,the positive expression of NF-κB,TNF-α, IKK-αmRNA were decreased evidently in the wolfberry + exercise group(P<0.05) in which the expression of IκB-α is the opposite tendency. The expression of TNF-α and NF-κBp65 is positive correlation, and IκB-α is negative correlation. Conclusion: 1. moderate-intensity aerobic exercise and black fruit wolfberry polysaccharides has Intervention in obstructive jaundice in rats, and can enhance obstructive jaundice induced liver injury in mice immune function, obstructive liver cells in mice with a certain repair. 2. Aerobic exercise and black fruit wolfberry polysaccharides in obstructive jaundice induced liver injury have significant therapeutic effects, its mechanism may be through inhibition of TNF-α-mediated NF-κB signaling pathway plays a key role.
