Objective: To study the effect of moderate-intensity aerobic exercise on the glucose metabolism of the rats with obstructive jaundice.Through the changes of PI3K/ AKT/ FoxO1 signal pathway,the mechanism of glucose metabolism and the mechanism of aerobic exercise were discussed.Methods:24 Kunming mice were randomly divided into three groups:control group(n = 8),model group(n = 8)and aerobic exercise group(n= 8).The model group and the aerobic exercise group were used to hang the total bile duct to the abdominal wall for 48 hours to construct the model of the obstructive jaundice.After 48 hours,the aerobic exercise group cut the fixed line of the common bile duct.After 7 days of normal feeding,no weight-bearing treadmill training was conducted.the adaptive training is carried out firstly,the medium-strength non-load-free running table training is adopted,the setting of the running platform gradient is 0percent,the training is carried out at a speed of 10 meters per minute,the training is trained for six weeks,the training is carried out for 60 minutes on a daily basis,and after 6 weeks,The mice were fasted on the previous night after the last training,and the following day was given anesthesia,and after the anesthesia was complete,the samples were taken,and the pathological changes of the liver tissue morphology and structure wereobserved with HE staining method.Blood glucose and total bilirubin TBIL were detected by blood biochemical test.The contents of AST and ALT were determined by Elisa.The liver PI3 K,protein AKT,FoxO1,G6 Pase were detected by Elisa.The content of glycogen synthase and PEPCK was detected by RT-PCR.The expression of FoxO1,PEPCK and G6 PasemRNA in liver tissue was detected by RT-PCR.Results:1.Serological results: Compared with the blank control group,the content of TBIL,ALT and AST in the model group was significantly increased(P<0.01);compared with the model group,the content of TBIL,ALT and AST in the exercise group decreased significantly(P <0.01);and compared with the blank control group,model group The blood glucose level of the mice was significantly higher(P <0.05).Compared with the model group,the blood sugar level of the mice in the exercise group decreased,but the difference was not significant(P> 0.05).The results of HE staining showed that the liver cells of the blank group showed a radial state.In the model group,the liver cells of the model group showed fibrosis,and the arrangement of the fibers was chaotic,the interval was widened,and the infiltration of the neutrophils in the liver and blood was increased;the liver fibrosis of the mice in the exercise group was improved,the interval was narrowed,and the structural change of the hepatic lobule was not large.3,the results of Elisa showed that: Compared with the blank control group,the content of liver glycogen in the modelgroup was significantly increased(P <0.01).Compared with the blank control group,the content of PEPCK and G6 Pase in the liver of the model group was significantly increased(P <0.01).Compared with the model group,the content of G6 Pase in the exercise group was significantly lower(P <0.05),and the content of PEPCK was significantly lower(P<0.01).Compared with the control group,the content of PI3 K,AKT and FoxO1 in the liver of the model group decreased significantly(P <0.01).Compared with the control group,the content of PI3 K,AKT and FoxO1 in the model group increased significantly(P <0.01).The mRNA expression of G6 Pase was significantly higher(P <0.01),and the mRNA expression of FoxO1 was significantly lower than that of the blank control group(P <0.01).Compared with the model group,the mRNA expression of PEPCK and G6 Pase in the exercise group was significantly lower(P <0.01).The expression of FoxO1 in the exercise group was significantly increased(P <0.01).Conclusion:(1)the model of liver injury induced by obstructive jaundice established successfully,and aerobic exercise can repair the liver injury caused by obstructive jaundice to a certain extent.After 6 weeks of successful modeling,glucose metabolism disorder appeared in the liver of mice with obstructive jaundice induced liver injury,while aerobic exercise could improve the disorder of glucose metabolism in mice with liver injury induced by obstructive jaundice.The mechanism is that aerobic exercise mayimprove the disorder of liver glucose metabolism by activating PI3K/AKT/FoxO1 signaling pathway.
