Seabuckthorn (Hippophae rhamnoides L.)is a widely used wild plant.Around the world our country is rich in sea buckthorn resource also Hippophae resources.As "Chinese Pharmacopoeia" noted Sea buckthorn has been used as medicinal and edible plant.Sea buckthorn fruit, leaves, seeds, flowers are rich in nutrients and bioactive substances, including sea buckthorn flavonoids which is the most unique and representative bioactive ingredient. clinical studies have shown that sea buckthorn flavonoids can lower blood pressure, blood sugar, anti-oxidation, anti-aging, antibacterial, anti-tumor, also enhance immunity, protect the liver, skin care, anti-allergic, anti-fatigue and other pharmacological activities, it can also effectively balance blood pressure, lower blood viscosity, inhibit excessive platelet aggregation, improve blood circulation, prevent atherosclerosis and so on. According to reports, the annual fruit marc of sea buckthorn industry generates up to one million tons after manufacturing, resulting waste of resources and destruction of the ecological balance. In this paper, using the QingHe County, Xinjiang sea buckthorn and optimize utilization of sea buckthorn marc exploration, mainly in the following research areas:(1) By studying single factor experiment, gaining the experimental factors and levels. Through orthogonal design method, optimize extraction conditions of total flavonoids of sea buckthorn from residues. The optimum conditions:50% ethanol concentration, extraction time 1.5h, solid-liquid ratio was 1:20, repeat three times. under optimum conditions, with five parallel experiments, validate the best extraction process. The results showed that under optimum conditions of sea buckthorn residues Total Flavonoids up 53.73 mg·g-1.(2) By studying the macroporous resin Static adsorption experiments, Compared of the adsorption properties of four different types of macroporous resin for total flavonoids of sea buckthorn residues, results showed that the most suitable for the purification of total flavonoids of sea buckthorn residues is D101 macroporous resin. Through dynamic adsorption experiments obtained by D101 macroporous resin purification of total flavonoids of sea buckthorn residues optimum parameters:sample concentration of 0.3145mg/mL, the sample flow rate 2BV/h, PH5. eluted with 5BV with the volume fraction of 70% ethanol to 2BV/h flow rate. further purification of total flavonoids sea buckthorn residues purity of 43.54 ± 0.32%(w/w), while the former purification of flavonoids purity was 21.64 ± 0.32%(w/w).(3) Through the soft capsule contents matrix dosage investigation, the best parameters institute of experimental suspending agents and wetting agents of the type and amount were below:(4) Sea buckthorn oil as a suspending medium, optimum mixing of sea buckthorn residues flavonoid extract powder with sea buckthorn oil ratio was 1:1.5, 4% beeswax as a suspending agent.(5) By using ultraviolet spectrophotometric method and HPLC method, sea buckthorn residues flavones soft capsule content of total flavonoids and isorhmnetin were determined. Rutin as reference substance, the sea buckthorn residues flavones soft capsule content of total flavonoids were determined.The results showed that sea buckthorn residues flavones soft capsule content of total flavonoids 31.02 mg/g. By using HPLC method, under the condition of Luna C18 (250×4.60mm,5?m) column as the stationary phase, methanol-0.4% phosphoric acid as the mobile phase flow rate was 1.0mL/min, column temperature 25?, the detection wavelength was 370nm, injection volume 10?L, measured as the average content of isorhmnetin 138.324mg/100g.(6) By using HPLC method, sea buckthorn fruit residues, fruit oil, nuts and flavonoids extract Powder isorhmnetin were determined. The results showed that Luna C18(250×4.60mm,5?m) column as the stationary phase, methanol -0.4% phosphoric acid as the mobile phase flow rate was 1.0mL/min, column temperature 25 ?, the detection wavelength was 370nm. isorhmnetin within 0.25?5.0?g/mL showed a good linear relationship;correlation coefficient of 0.9999.The average recovery was 100.9%.Measured of sea buckthorn fruit residues?nuts?flavonoids extract Powder?fruit oil as the average content of isorhmnetin was 95.35?105.77? 191.87?17.27 mg/100g.
