Paprika oleoresin is a high-quality natural pigment in the red pepper,which is also known as red pepper pigment,capsaicin.It is an end product of carotenoids synthetic pathway in red pepper.Paprika oleoresin is a kind of sweet smell chili dark red oily liquid and usually exists as the mixture of capsaicin.The paprika oleoresin is widely used in food,food additives,cosmetics,feed and other fields due to its property of bright color,strong color given,thermostability,safe and reliable and so on.Therefore,it is a study of significance about paprika oleoresin extraction technology in improving the added value of pepper resources and improving the depth of agricultural development and utilization.In this study,supercritical carbon dioxide extraction technology was used to extract paprika oleoresin form red pepper.And the in vivo and in vitro antioxidant properties of paprika oleoresin were studied,to get theory basis for utilizing paprika.The research aimed to study the effects of extraction pressure,separation time,extraction temperature and separation temperature on the extraction rate and color scale.As a result,the optimum conditions were obtained.After,effects of the paprika oleoresin on hemolysis of rat RBC,rat liver homogenate and MDA formation in rat liver mitoehondria in vitro were studied.The main results were summarized as follows:The results showed that the optimum conditions of supercritical CO2 fluid extraction were:the particle size of 60 mesh,the separation temperature of 50 ?,the extraction temperature of 40 ?,the extraction pressure of 20 MPa,CO2 flow of 20 L/h and extraction time of 3 h.In this condition,color value was 173.8 and the extraction rate was 4.29%.Hemolysis of rat RBC,rat liver homogenate,MDA formation of rat liver mitoehondria were inhibited in a dosage-dependent manner by the paprika oleoresin in the tested concentration of 0.5-10 mg/mL.The results showed that paprika oleoresin had antioxidant activity in vitro.With the increasing of the dose,paprika oleoresin could increase the capability of SOD and GSH-Px activity in serum of S180 tumor-bearing mice,reduced the level of MDA and thus improved the activity of antioxidant capability in vivo.When dose reached 300 mg/kg/d,paprika oleoresin was no significant differences with 5-Fu of 10 mL/kg/d for improving the capability of antioxidant of tumor-burdened mice in vivo.
