Ganoderma lucidum is a mushroom having been widely used in China as Traditional Chinese Medicine as well as healthy food for thousand years,which could promote longevity and health.The polysaccharides,which are the main active components of G.lucidum,are capable of anti-oxidation,anti-aging and immunoregulation.In this paper,the components and the in vitro antioxidant activities of crude polysaccharides of Ganoderma lucidum from different origins were studied.Then,the extraction technology and the structure characteristics of the polysaccharide of Ganoderma lucidum were studied,and main results were as follows:Firstly,the components of Ganoderma lucidum from different origins were investigated.The result indicated that the content of some functional components(total carbohydrate,energy,Ge,protein,total amino acid,essential amino acid,ganoderic acid A,B,et al)of An Hui no.1 sample was higher than the content of some functional components of Ji Lin no.1 sample and Ji Lin no.2 sample.ABTS radical scavenging activities,DPPH radical scavenging activities,hydroxyl radical scavenging activities and K3Fe(CN)6 reduction methods were employed to compare the in vitro antioxidant activities of three crude polysaccharides of fruit bodies from different origins and one exopolysaccharide of liquid fermentation.The results showed that four different crude polysaccharides had certain abilities of antioxidant activities.The crude polysaccharide of An Hui no.1 sample showed the strongest capacity for scavenging ABTS and DPPH radical with EC50 values of 1.50 mg·mL-1 and 2.09 mg ·mL-1,respectively.Besides,the crude polysaccharide of An Hui no.1 sample showed the most potent capacity for reducing activity.The crude exopolysaccharide of liquid fermentation showed the most potent capacity for scavenging hydroxyl radical with EC50 value of 1.29 mg·mL-1.It was concluded that the antioxidant activities of four crude polysaccharides were different.Moreover,the crude polysaccharide of An Hui no.1 sample showed the strongest antioxidant activity.The extraction process of the polysaccharide of An Hui no.l sample was optimized through single factor experiments and RSM design tests.The optimal conditions of polysaccharide were obtained as ratio of liquid to material 21:1,extraction time 2.5 h,extraction temperature 100 ?,extraction number 3.Under these optimal conditions,the yield of polysaccharides was 1.54±0.04%closed to the predicted value 1.52%.GLP-1,GLP-2 and GLP-3 were obtained by the means of purification using DEAE-650 ion exchange resin and gel sephadex G-100 successively.The results of FTIR,UV spectrum and basic physical and chemical properties tests showed that GLP-1,GLP-2 and GLP-3 had the characteristic absorption peaks of polysaccharides,and there were no protein,nucleic and pigment in them,and they had the general characteristics of polysaccharides.HPLC pre-column derivatization analysed its monosaccharide composition.The results showed that GLP-1 was composed of mannose,glucose,galactose,xylose and fucose.GLP-2 and GLP-3 were composed of mannose,ribose,rhamnose,glucuronic acid,glucose,galactose,xylose and fucose.
