Ganoderma Lucidum is one of the most famous traditional Chinese tonics and drugs. Selenium is an essential trace element for human beings.The researchs on the activites of Ganoderma Lucidum and selenium have been deeply performed,but studies on the use of Ganoderma Lucidum to biotransform selenium have not been repored overseas and seldom reported domestically.Polysaccharide containing selenium in Ganoderma Lucidum is made of both natural and artificial synthesized Fungi polysaccharide enriched selenium,in which inorganic selenium in the medium can be transformed into organic selenium by Ganoderma Lcuidum.Research results have showed that polysaccharide containing selenium in Ganoderma Lucidum has better pharmaceutical activities than polysaccharide or selenium.therefore,it is more practical to study Se-polysaccharide in Ganoderma Lucidum.The results of this study were as follows:This study compared the Se-riched capacity and the conversion rate of organic Se of Ganoderm lucidum cuturedon amone Se-riched liquid culture fermentation,mixed fermentation and Se-riched solid-state fermentation.The effect of selenium on production and the structure of polysaccharide of Ganoderm lucidum was also be studied.The results here show that content of organic Se of mycelium can be achieved at 6333.59μg/g on Se-riched liquid culture fermentation when the concentrate of Se is 500μg/mL;when is 700μg/g,the maxium Conversion rate of organic Se can reach at 18.58%on Se-riched solid-state fermentation.When the concentrate of Se is below 200μg/g on solid-state fermentation,the production of polysaccharide can be improved.The analysis of IR spectrum of polysaccharide of Se-riched G9,revealed that Se did not change the main structure of polysaccharide of G9 and speculated that Se may be being as selenium ester (Se-OH).Through the comparation between Se-polysaccharide and polysaccharide on scavenging effect and reductive capacity,we found that the Se-polysaccharide is better than polysaccharide with 95%scavenging rate in concentration of 10 mg/mL and also a better reducer in the same concentration.This showed that the Se-polysaccharide have better effect on antioxidant activity out of body.Sixty KM male mice were divided in 5groups randomly by weight inchuding control group,medium dose of polysaccharide group(100 mg/kg·d),high dose of Se-polysaccharide group(200 mg/kg·d),medium dose of Se-polysaccharide group(100 mg/kg·d),low dose of Se-polysaccharide group(50 mg/kg·d) and 12 mice in each group.During the experimental session,the 5 test groups were administered polysaccharide solution every day by 30 days and the control group was administered the normal saline.After 30 days,we tested the superoxide dismutase(SOD),Glutathione peroxidase(GSH-PX) and malomdialdehyde(MDA).The results showed that the Se-polysaccharide can improve the activity of SOD and GSH-PX obviously,also can reduce the content of MDA in mice liver tissue to some extent.So,the Se-polysaccharide has the antioxidant activity.The research futher improved the method of determinating polysaccharide of Ganoderma Lucidum.We usesα-amylase to hydrolyzate starch which is attached on the myceil of Ganoderma Lucidum and study the process of hydrolyzation of amylase during the process of extracting the polysaccharide.The best processes of hydrolyzation were that determined by single-factor test and orthogonal test.The results showed that the enzyme concentration,time and temperature of hydrolyzation were 0.46μ/mL,20 min, 50℃.The researches on method of using phenol-sulphuric acid to detect the content of polysaccharides have done.The best method was etermined by single-factor test and orthogonal test.The results showed that the best way is 2.0 mL test solution adding to 1 mL 5%phenol and 5 mL sulphuric acid,then standing in 20℃for 30 min and determinating at 490 nm.
