Chemiluminescence analysis is a trace analysis method which is based on the some point of the light-emitting quantum or total luminous intensity to quantitative analysis of the material under test.Chemiluminescence analysis method has many advantages including accurate and easy operating,high sensitivity and simple equipment.Luminol-H2O2 CL reaction,as a popular CL reaction,has been widely applied for the determination of H2O2 and glucose.The CL signal of luminol-H2O2 is quite weak in the absence of catalyst?The CL reaction of luminol-H2O2 can be catalyzed by several kinds of catalysts including metal ions,metal nanoparticles(NPs),and enzymes.Among them,horseradish peroxidase(HRP)has been widely used to fabricate sensors for H2O2 and glucose.However,natural enzymes suffer from some serious disadvantages such as rapid loss of activity,expensive and time-comsuming preparation and purification.Therefore,looking for more excellent catalyst or sensitization agent research is of great significance.In this study,we found that iodophenol blue and AgCNs can catalyze the luminol-H2O2 CL reaction and enhance the week chemiluminescence(CL).The proposed method has been applied to the detection of glucose in diluted serum.The results of CL detection of glucose were in good agreement with those obtained by a commercial paper-based method.The result is sactisfactory.The research in my assay consists of two parts mainly:Chapter 1: Indophenol-blue-enhanced Luminol chemiluminescence and its application in hydrogen peroxide and glucose detectionIn this study,we exploreded the catalytic activity of iodophenol blue on the luminol-H2O2 chemiluminescence(CL)system which is a classical model CL system.Step one,luminal-H2O2 CL system which was catalyzed by iodophenol blue was established and preliminary explored the mechanism that iodophenol blue catalyzed the luminol-H2O2 system.The second step,hydrogen peroxide was regarded as the analyte,the luminol-H2O2-iodophenol blue system was applied to quantitative analysis of hydrogen peroxide.Under the optimum experimental conditionals,a linear relationship between the relative CL intensity and H2O2 concentration in the range of 0.025-10 ?M was obtained.As low as 14 nM H2O2 can be sensitively detected by using the proposed method.Finally,we applied the proposed method to determinate the glucose in human serum.Experiment results showed that there is a good argreement between the proposed CL method and the glucose paper method,indicating the capability of the proposed CL method being applied to the determination of glucose in biolgical sample with satisfactroy result.Chapter 2: AgCNs-enhanced Luminol chemiluminescence and its application in hydrogen peroxide and glucose detection.This chapter is based on silver nanoparticles cluster catalyzed luminol-hydrogen peroxide CL system,building the high sensitivity detection of hydrogen peroxide and glucose analysis method.Under the optimum experimental conditions,the linear range of detection of hydrogen peroxide is 0.025-75 ?M,the detection limit of detecting hydrogen peroxide is 1.7 nM.The linear range of detection of glucsoe is 0.01-50 ?M,the detection limit of detecting hydrogen peroxide is 7.3 nM.
