Extraction Optimization And Hypoglycemic Lipid-lowering And Antioxidant Effects Of The Efficacy Components In Doellingeria Scaber

Taking the stem and leaves of Doellingeria scaber as raw material,extracting total flavonoids,total saponins and total polysaccharides from Doellingeria scaber with Ultrasonic method and the extration conditions were screened by Design-Expert8.05 b software to get the best extraction conditions,and to investigate the hypoglycemic,hypolipidemic,anti diabetic mice of oxidation.The results are as follows:1.By ultrasonic method on the effective components of flavonoids extraction optimization of Doellingeria scaber experiment,with the extraction of crude flavonoids rate as the index,the results showed that liquid ratio was 39.27:1,the ethanol concentration was 71.09%,ultrasonic time were 62.44 mins.The extraction rate of flavonoids was 1.39%.2.In ultrasonic assisted extraction of saponin from Doellingeria scaber in the optimization of extraction test,extration rate of crude sapoin as evaluation indexes,the results showed that liquid ratio 45.17:1,ultrasonic temperature was 51.05?,the ultrasonic time were 61.19 mins.The optimal extraction ratio was 1.74%.3.In the ultrasonic-assisted water extraction optimization test on the effective components of Doellingeria scaber polysaccharide,extraction of crude polysaccharide ratio as index,test results showed that the ratio of material to liquid 60.24:1,ultrasonic temperature is 51.53?,the ultrasonic time was 93.76 min.The maximum extraction rate was 4.16%.4.In the test group of reducing blood glucose in diabetic mice by total flavonoids,Doellingeria scaber total saponins and polysaccharide extracts were detected in diabetic mice,fasting blood glucose and glucose tolerance.I recorded the amount of water and food intake and body weight transform.The experimental results indicated that three kinds of ingredients could reduce the fasting blood glucose(Glu),improved glucose tolerance,and effectively alleviate the symptoms,Doellingeria scaber total saponins,total flavonoids effect effect of high dosage group and positive control group.The effect of high dose group was the second.5.Taking the extracts of total flavonoids,total saponins and polysaccharide of Doellingeria scaber as experimental group,we tested the blood lipid indexes of the diabetic mice.The experimental resulted showed that the three kinds of food ingredients can effectively adjust the Doellingeria scaber TC,TG,LDL-C,HDL-C content in serum,the effect of total saponins of Doellingeria scaber the best.6.Taking the extracts of total flavonoids,total saponins and polysaccharide of Doellingeria scaber as experimental group,we tested the Antioxidant indexs of the diabetic mice.The experimental results showed that three kinds of ingredients could reduce the content of liver tissue in MDA,GSH-PX,CAT,increase the content of SOD,enhanced its the total saponins of antioxidant capacity,the effect of the best Doellingeria scaber.
