Pleurotus(Fr.)Kumm,a kind of wood-roting fungi,is easy to cultivate and delicious to eat.It has high economic value and medical benefits,which is also closely related to human life.Pleurotus abalonus belongs to Pleurotus,which is a kind of income fungus in southern Shaanxi.As the main varieties of fungus,Pleurotus is abounding in southern Shaanxi,but the species of Pleurotus are often confused.In order to avoid species confusion effectively,and establish a theoretical basis of Pleurotus germplasm resources,the genetic diversity of Pleurotus was evaluated in southern Shaanxi.Furthermore,optimizing the liquid fermentation medium of Pleurotus abalone mushroom can provide a theoretical reference for the pilot production of new strains of Pleurotus in southern Shaanxi,which could be helpful in expanding the cultivation and production of new strains.In this study,35 strains of Pleurotus from southern Shaanxi were adopted.Genetic diversity among different strains was studied according to the morphological characters,antagonistic reaction,Esterase Isozyme and ITS sequence.The liquid fermentation medium of Pleurotus abalonus was also studied by response surface methodology.1.The biological characteristicsThe agronomic traits(cultivation methods,temperate,fast-growth,anti-hybrid etc.),colony morphology(colour,growth rate,density etc.)and fruiting body morphology(cap colour,shape,biomass,etc.)of 35 Pleurotus strains were studied by the method of tissue culture and cultivation test.The results show that the biological characteristics are rich in the test strains.According to colony morphology in the solid culture conditions and the degree of antagonistic reaction,they can be classified into 5 categories and 4 categories respectively.Moreover,according to the fruiting body morphology,they can obviously be divided into C23(light red fan-shaped cap),C27(yellow funnel cap)and C29(light yellow hemispherical cap)(not Pleurotus),and the C4 mycelium and fruiting body has the faster growing speed,high biological conversion rate,anti-noise ability,so the advantage of cultivation is significant.2.The genetic diversityDifferent species and genetic relationship of the 35 strains were identified by studying the Esterase Isozyme and ITS sequence,which is also combined with the biological characteristics.The results show that there are 9 strains of Pleurotus floridanus,8 strains of Pleurotus eryngii,7 strains of Pleurotus ostreatus,6 strains of Pleurotus sapidus,1 strain of Pleurotus nebrodensis,1 strain of Pleurotus abalonus,1 strain of Pleurotus salmoneostramineus,1 strain of Pleurotus citrinopileatus and 1 strain of Flammulina velutipes(not Pleurotus).Furthermore,there are several homonyms such as C2 and C11,C17 and C18,C19 and C20,C21 and C25.In addition,C23,C27 and C28 have different names,could be different species.3.The liquid fermentation medium of Pleurotus abalonusThe suitable liquid fermentation medium of Pleurotus abalonus mushroom were optimized by the Plackett-Burman test and the steepest ascent test,Box-Benhnken response surface method also included.The best fermentation medium are as follows(g/L): potato 240.00,glucose 30.00,corn powder 15.00,peptone 5.00,KH2PO4 3.00,MgSO4·7H2O 2.00,VB1 0.10,1 L of water,natural pH.On these conditions,the mycelium has small particles and high density,mycelium biomass of Pleurotus abalonus mushroom is up to 15.126 g/L,which was increased by 17.92% compared with the initial condition.
