| Lycium ruthenicum Murr which is distributed in the western desert areas of China’s wild plants,belongs to the genus Lycium of the family Solanaceae.The L.ruthenicum is sweet and natured,an effective natural antioxidants,it’s rich in vitamins,minerals and other nutrients content,not only can be used as medicine,but also windbreak and sand fixation,control saline land.L.ruthenicum products mainly come from wild,in recent years,due to the deterioration of the growth environment and people over harvesting,the resources have been damaged to a certain extent.L.ruthenicum seeds of conventional seedling germination rate is low.Therefore,investigate the characteristics of seed germination and tissue culture so as to achieve the purpose of rapid propagation,which has a great significance of protecting and developing L.ruthenicum resources.In this experiment,using L.ruthenicum seeds as material,through(1)Determination of water absorption rate of seeds;(2)Effects of soaking temperature on seed germination;(3)Effects of hormone on seed germination;(4)The callus induction of L.ruthenicum;(5)Induction of adventitious buds;(6)rooting induction;6 aspects to study germination characteristics and tissue culture methods of L.ruthenicum.The main conclusions are as follows:1.The former 2 h is the stage of fastest rapid water absorption,2-4 h the rapid water absorption become slower,after 8 h tends to be stable,and the overall water absorption rate is fast.2.Soaking seeds at low temperature under 10 ℃accelerated the germination process of seeds and improved the germination rate.Compared the germination of germination bed by filter paper and MS medium,filter paper is better.10 day was suitable for the germination vigor of L.ruthenicum seeds.3.Hormone treatment can effectively improve the germination rate of seeds,300mg·L-1GA3,,100 mg·L-1NAA,300 mg·L-1IAA,200 mg·L-16-BA soak the seeds 24 h canincrease germination rate effectively,GA3 is the best.4.According to the different concentrations of 6-BA and NAA ratio,time statistics of callus,the callus induction rate and growth,the results show that the MS +0.1 mg·L-1NAA 1mg·L-16-BA induced callus growth best,callus showed fast and healthy dense green clumps,without vitrification,the highest rate reached 95%.5.By comparing the different concentrations of 6-BA,the time of adventitious bud formation,the induction rate and the growth rate were observed.The results showed that the callus differentiation induced by 0.2 mg·L-16-BA concentration was the best.6.By comparing the different concentrations of IBA,statistics the rooting time,rooting rate and growth potential,the results showed that 0.2 mg·L-1lIBA induced rooting of clustered shoots was the best. |
