This research established the rapid propagation system of the late-flowering chrysanthemum (HR-1) by combining the technology of tissue culture and cutting propagation. The study plants of HR-1 were selected by irradiation mutation formerly and were expected to grow to gorgeous blossom and abundant foliage that could be for sale during the Spring Festival based on this technology. The main results are as follow:1. This research established an efficient tissue culture system of HR-1 chrysanthemums by leaves culture. The experiment mainly included three parts: callus induction, bud differentiation and root induction. (1) The experiments of callus induction were conducted by using the HR-1 leaves as explants with the MS as the basic culture medium, and adding the exogenous growth regulator combination as 6-BA (1.0?3.5 mg/L)+NAA (1.0?2.0 mg/L). Results showed that the most effective condition is using the 6-BA (2.5 mg/L)+NAA (1.0 mg/L).In this case, the callus induction rate can reach 96.6%, and the callus induced are fresh and in tender green that growth quickly and favorite for following induction; (2) The bud differentiation induction was carried out by using the above callus as material, the MS as the basic culture medium, and adding the exogenous growth regulator combination as 6-BA (1.0?3.5 mg/L)+NAA (0.0?0.2 mg/L). We find that 6-BA (3.0 mg/L)+ NAA (0.1 mg/L) had best effects and the bud differentiation rate can reach 93.3%. (3) Using the above bud with 1/2 MS as the basic culture medium and adding the exogenous growth regulator combination as 6-BA (0.0?0.2 mg/L)+NAA (0.0?0.2 mg/L), the root induction and differentiation was conducted. Result showed that no exogenous growth regulator with 1/2 MS medium or 1/2 MS+NAA (0.1 mg/L) had best effects. The average of each differentiation bud can take 10.48 roots.2. In order to speed up the marketing of the flower, we applied the cutting propagation combined with tissue culture, i.e. conducting the cutting propagation in the vegetative stage of the cultured plantlets. The experiments of cutting propagation of HR-1 mainly included three parts:the effect of matrix, the management of the cutting, planting time on the survival rate and flowering stage of HR-1 chrysanthemum cuttings. (1)For studying the effect of substrate on the survival rate of cutting seedlings, we cut 8.0 cm long HR-1 branches of outdoor planting seedlings which grew well from the test tube seedlings, and soaked the branches in NAA at 1.0%(W/V) for 2 hours, and inserted in three kinds of matrix. The results showed that the survive rate of cutting seedlings was highest as 93.3% in the matrix of the peat 60%, vermiculite 20% and garden soil 20%. (2) We studied the survival rate of different cuttings propagation under two kinds of management, open and filmed nurse. Results showed that the survival rate under filmed nurse is higher than that under open management. The seedling survival rate is as high as 90% by filmed nurse when cutting with 30 days; (3)In order to make the HR-1 chrysanthemum can be gorgeous blossom and abundant foliage, and may be for sale during the Spring Festival, we carried out three cutting time experiments, including April 25, June 15 and September 15. The results showed that HR-1 chrysanthemum which cuttings in June 15th can be gorgeous blossom and abundant foliage, which is suitable for sale during the Spring Festival. (4)Topdressing compound fertilizer is given about within 15 to 30 days a time. The watering depends on the soil moist degree in look as dry and wet.In this study, the rapid propagation technology system of HR-1 chrysanthemum was established by combining tissue culture with cutting propagation. It provided a new way for the rapid propagation of the mutation breeding technology of flowers, and have good marketing value.
