Angelica sinensis(Angelica sinensis(Oliv.)Diels)is one of the varieties of authentic Chinese herbal medicine in Gansu province.The annual output of more than 90% of the total production of the country,with and blood and other functions,such as the dosage of medicinal herbs increased year.However,with the prolongation of planting years and the aggravation of global warming trends,the disease degree of Angelica seedlings increased,the early bolting rate was raised,which caused the seedling areas to change to higher elevation,seriously destroyed the ecological environment.Effected the sustainable development of Angelica production,but using tissue culture method to reproduce,because the seed germination rate is very low.It is difficult to achieve seedling breeding.Aiming at the above-mentioned problems,according to the principle of crop micro-ecology.The study objects of Angelica sinensis seed breeding in Zhang County,Gansu Province,this paper systematically studies the germination characteristics of Angelica seeds and the distribution of endophytic flora in seedling roots,aiming to explore the treatment methods for improving the germination characteristics of Angelica seeds,revealing the micro-ecological characteristics of Angelica.The providing scientific basis for formulating the regulation rules of Angelica.Results are summarized as follows:1.Different concentrations of Hg Cl2 solution,(1.0 mmol/L,1.5 mmol/L and 2.0mmol/L)soaking different time(0,1,3,5 and 7 min)have a very significant effect on the germination rate of Angelica seeds,with no treatment of dried seeds for control.With no treatment of dried seeds for control,and 1.0 mmol/L immersion 5 min for best treatment,followed by 3min soaking.The optimum concentration and processing time can significantly improve the germination characteristics of Angelica seeds and improve the resistance of seedlings,and lay a good physiological and biochemical basis for strong seedlings.2.The solution soaking with different time(ck,T1,T2,T3,T4 and T5 h)has a significant effect on the germination characteristics of Angelica seeds,including T2 h as the most suitable soaking time,followed by soaking T3 h.Suitable time has promoted effect on seed germination of Angelica.3.The Study of endophytes on the separation of seedlings from Angelica showed that Angelica seedling roots in isolation and purification of the 29 strains of endophytic bacteria,of which 20 strains for endogenous bacteria and 9 strains for endogenous fungi,which which the fungi contains 4 strains mold and 5 strains of yeast.4.To extract angelica root after separation and purification of bacteria,with the molecular identification of 16 s r RNA,after extraction of plasmid,sequencing,by using NCBI databases,there were budding identified Bacillus sp.,Pseudomonas argentinensis,Devosia neptuniae,Variovorax sp.,Brevibacterium sp,Pseudoxanthomonas sp.,Rhizobium nepotum,Sphingobium sp.and Microbacterium sp.for 9 kinds of endophytic bacteria,which the obvious advantage for Bacillus strains.
