| In recent years,along with the rapid development of the Lentinus edodes industry in Hanzhong,it has brought about economic benefits while bringing a series of problems such as environmental pollution.About among them,the most prominent problem is how to dispose of a large amount of edible fungus waste materials.In this study,a relatively high nutritional value but relatively scarce,less research Dictyophora as the test material,using waste materials as the main raw material to select the best cultivation medium and production media formula,using plastic turnover box and turnover box the factors influencing the growth of Dictyophora were screened,and improving Dictyophora cultivation techniques with raw materials and covering soil in greenhouses.While reducing the production cost of Dictyophora,it also achieved the goal of resource utilization of edible mushroom farmer cultivation waste materials in the Hanzhong area.The results of the experiment are as follows:1.Nutrition and environmental conditions optimization of the Dictyophora"Jiyou 1".The single factor test was used to screen the nutrient conditions and environmental conditions of the Dictyophora’s stock culture:The optimum carbon source was corn powder 20 g,nitrogen source5 g soybean powder,C/N ratio 20:1,inorganic salt MgSO4 0.5 g,growth the factor is VB1 0.5g;the optimum temperature range is 2628℃,the optimal temperature is 28℃,the optimum pH range is 4.56,and the optimal pH value is 4.5.Orthogonal test was used to screen the best medium for the Dictyophora’s stock culture:22 g corn powder,4 g soybean powder,0.5 g MgSO4,0.5 g VB1,1.0 g KH2PO4,8.015.0 g agar and 1000.0 mL water.The single factor experiment was used to screen the nutrient conditions and environmental conditions of the Dictyophora’s liquid species:The optimum carbon and nitrogen source and inorganic salt were corn powder 20 g,soybean powder 5 g,and MgSO4 0.5 g;the optimal incubation days were 12d and the rotation speed was 110 r/min,optimum temperature range 2428℃,optimum culture temperature 26℃.Orthogonal test was used to screen the best medium for the liquid medium of Dictyophora:22 g corn powder,4 g soy flour,MgSO4 0.4 g,KH2PO4 1.0 g,and 1000.0 mL water.2.Analyze the nutrient composition of waste materials of edible fungus,and screen the cultivated species and production medium formulation of the Dictyophora.The use of ICP-OES,kjeldahl method and potassium dichromate external heating method to determine whether the carbon-nitrogen ratio and trace element content of the edible fungus waste used in the test satisfies the growth requirements of the Dictyophora.The results show that the content of Ca in the waste material was 13.6 g·kg-1,K=5.60 g·kg-1,P=4.83 g·kg-1,S=4.72 g·kg-1,Mg=3.98g·kg-1,Na=1.23 g·kg-1,and the content of other elements was less than 1.23 g·kg-1.The carbon content of the waste material was 62.6%,and the nitrogen content was 2.6%.The single-factor test was used to screen out the best medium for cultivation of Dictyophora"Jiyou 1":Waste material 75%,bran 20%,soybean powder 3%,gypsum powder 1.5%,sucrose 0.5%,and The amount of water is 60%;the optimum production medium formulation is 85%of waste materials,8%of mixed wood chips,8%of wood chips,5%of rapeseed rods,1%of superphosphate,1%of gypsum powder,and 60%of water content.3.Screening of factors affecting growth of Dictyophora:The growth range of hyphae was2226℃and the optimum temperature was 24℃.Fruiting stage:the optimum growth condition was 26℃at 2228℃;the best effect of air relative humidity was 80;the normal growth range was 4.56.0,and the optimum pH was 5.0;the best covering material was humus.4.Greenhouse raw material cover soil cultivation techniques:Using greenhouse cultivation techniques,comparing the production medium D3 and the traditional cultured Dictyophora culture medium E in the yield and bioconversion rate of the Dictyophora,the test results production medium D3 has obvious effect of increasing production and income,the output of Dictyophora reaches 162.30 kg·500m-2,which is nearly 10%higher than that of traditional cultured Dictyophora culture medium,and the bioconversion rate reached 47.12%. |
