Objective: This study aims to optimize the extraction method of three active ingredients phenolic acids, flavonoids and anthraquinones from Japanese ampelopsis, and to quantify these three active ingredients using gallic acid(GA), quercetin and emodin as standards.Methods: On the basis of single factor investigation, the concentration of extraction solvent, time, temperature, liquid ratio and other related factors were optimized by orthogonal experiment, which provided the optimized conditions for extracting three active ingredients of Japanese ampelopsis. High performance liquid chromatography(HPLC) method was used to analysis the content of its main components.Results: 1.phenolic acids: the highest extraction yield of phenolic acid compounds was achieved by using ultrasonic-treated organic solvent. The parameters of this method are as follows: methanol concentration is 60%, extraction temperature is 80 oC, extraction time is 30 min and solid-liquid ratio is 1:20. The extraction efficiency of phenolic acid was 0.782% under this optimized conditions.2.flavonoids:The most efficient extraction method for quercetin extraction was methanol-hydrochloric acid extraction method. The parameters of this method are as follows: methanol-hydrochloric acid ratio is 6:1, extraction time is 30 hours and solid-liquid ratio is 1:40. The extraction efficiency of meletin was 0.9922mg/g under this optimized conditions.3.anthraquinones: The parameters of extraction method that produced the highestconcentration of emodin are as follows: ethanol concentration is 95%, extraction time is30 min and solid-liquid ratio is1:10. The extraction efficiency of emodin was 0.631% under this optimized conditions.Conclusion: 1.phenolic acids: Application of ultrasonic-treated organic solvent yields the highest percentage of target ingredient from Japanese ampelopsis. 2. flavonoids: The highest extraction rate of quercetin from Japanese Ampelopsis was obtain by using methanol-hydrochloric acid extraction method. 3.anthraquinones: The best extraction method of active ingredient emodin from Japanese Ampelopsis was determined by orthogonal experiment.
