Objective:In this paper,to study the quality standards of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn,it was researched that the processing technology of carbo Nelumbo nucifera Gaertn and the changing law of the main chemical composition of Nelumbo nucifera Gaertn fried carbon before and after.This will provide a reference for the establishment and perfection of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn quality standards,in order to ensure the use of two pieces safe,effective,stable and controllable.Besides,the pharmacodynamics of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn were processed a preliminary study.Methods:1.To study the processing technology of carbo Nelumbo nucifera Gaertn,the contents of quercetin in Nelumbo nucifera Gaertn was taken as index and frying time and temperature taken as the effects.The orthogonal design method was used to carry out the optimization of the processing technology of carbo Nelumbo nucifera Gaertn;2.The contents of alkaloids,quercetin,iso-quercetin and hyperoside in Nelumbo nucifera Gaertn were determined by HPLC,while researched a comparative study of the content changing of over several components processed before and after; The variation of alkaloids,quercetin,iso-quercetin and hyperoside in Nelumbo nucifera Gaertn was studied by an analog processing method;3.According to quality requirements of pieces in the 2010 edition of "Chinese Pharmacopoeia",combined with the difference of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn,established quality standards in terms of identification,examination, determination,etc. 4.SD rats were used as experimental animals,and administrated by the extract of different processed products;Selected bleeding time,clotting time,thromboplastin time APTT of activated partial,prothrombin time PT and thrombin time(TT),etc as indicators to study the changing of blood coagulation of Nelumbo nucifera Gaertn processed before and after.Results : 1.The optimum processing conditions of carbo Nelumbo nucifera Gaertn were determined:fried at 280? for 12 minutes by the orthogonal design method with taking quercetin as the index; 2.With the contents determination of alkaloids,quercetin,iso-quercetin and hyperoside in Nelumbo nucifera Gaertn by HPLC,the results showed that there were different main chemical ingredients in Nelumbo nucifera Gaertn from different areas;The alkaloid content of carbo Nelumbo nucifera Gaertn wassignificantly reduced,almost wiped out;The hyperoside and iso-quercetin content also reduced significantly when Nelumbo nucifera Gaertn was fried carbo,and decreased gradually with extending processing time,finally almost wiped out;The quercetin content showed a sharp change:increased at first then decreased with extending processing time.The results of simulate experiment showed that the increasing of quercetin was related with thermal decomposition of hyperoside and Isoquercitrin;3.The spots were clear and good resolution on TLC;It was maked limitation the moisture, ash, acid insoluble ash,extract,the main active substance content,etc in Nelumbo nucifera Gaertn and its processed products.4.In the aspect of coagulation tests,raw Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn could shoten bleeding and clotting time of SD rats compared with blank group,and it enhanced after Nelumbo nucifera Gaertn was fried carbo;Compared with the control, raw Nelumbo nucifera Gaertn could shorten rat PT(P<0.05),the impact of APTT and PT was significant that rats were administrated carbo Nelumbo nucifera Gaertn(P<0.01),carbo Nelumbo nucifera Gaertn could also shorten rats TT(P <0.05),the impact of TT in the other groups were no difference; Compared with the raw Nelumbo nucifera Gaertn group,the impact of rats APTT,PT,TT in carbo Nelumbo nucifera Gaertn group had difference(P<0.05).Conclusion:The experiment determined the processing technology of carbo Nelumbo nucifera Gaertn.It was explored through the method of simulate experiment that the content variation of the chemical constituents and the processing mechanism when Nelumbo nucifera Gaertn was fried carbo.On the basis of the study of quality standards of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn,established the TLC conditions to identify Nelumbo nucifera Gaertn;Coagulation experiment showed that carbo Nelumbo nucifera Gaertn had a role in blood clotting,and the blood clotting mechanism was a result of endogenous and exogenous action.The study will provide preliminary theoretical and experimental basis for the quality control,the clinical application and the further research of Nelumbo nucifera Gaertn and Nelumbo nucifera Gaertn processed products.
