Aim The aim of this study was to examine serum betatrophin concentrations, plasma glucose, insulin, lipids and other indicators in patients with type 2 diabetes and normal plasma glucose population, and analyze the correlation of betatrophin concentrations with insulin resistance and islet beta cell function, and the relationship between serum betatrophin levels with lipid. On this basis to investigate the clinical value of betatrophin on human glycolipid metabolism, and provide a basis for its clinical application in the future. Methods The study included 78 the fasting plasma glucose were normal according of glucose tolerance test results.according to the fasting insulin(FINS)levels non-diabetic group was divided into two groups,FINS < 12.0m IU/L as control group(NC group,n=49),FINS ?12.0 m IU/L for hyperinsulinemia(HINS group n=29).and select the type 2 diabetes hospitalized in the department of endocrinology(T2DM)patientsas(T2DM group n=33).The questionnaire was used to investigate the basic clinical indexes of the subjects, and the levels of betatrophin were determined by enzyme-linked immunosorbent assay, and the levels of 25-OH-VD were measured by radioimmunoassay.SPSS17.0 software was used to analyze and compare the difference of each index and the correlation between serum betatrophin levels and each index? Results 1.Serum betatrophin levels were elevated in theT2 DM group compared with the NDM group.T2 DM group,NC group,HINS group showed a downward trend.Serum 25-OH-VD levels in T2 DM group were significantly lower than those in NDM group.2.Correlations of all indicators with serum betatrophin concentrations.Serum betatrophin levels were positively correlated with age, waist to hip ratio,TCH and LDL,and negatively correlated with BMI,hipline,FINS,HOMA-IR,HOMA-? and 25-OH-VD.Serum betatrophin levels were not correlated with sex, weight,TG,HDL,FPG.Circulating betatrophin concentrations was positively correlated with age,but negatively correlated with BMI,FINS,HOMA-IR,and HOMA-? in NDM subjects.Multiple linear stepwise regression analysis: age and 25-OH-VD levels were independent factors of betatrophin.Serum betatrophin levels were associated with age in NDM subjects.3.Comparison of clinical indexes between groups. There were no significant differences in gender, height, diastolic blood pressure,AST and HDL among the NC group,HINS group and T2 DM group.Systolic blood pressure and FPG were significantly higher in the T2 DM group. The TCH and LDL levels were significantly higher in group T2 DM.The weight,BMI,hipline, and FINS levels in the HINS group were significantly higher than those in NC group and T2 DM group,HOMA-IR and HOMA-? had significant difference among the 3 groups,HOMA-IR decreased gradually in HINS group,T2 DM group,NC group presentation,HOMA-? decreased gradually in HINS group,NC group,T2 DM group.4.Correlation analysis showed that there was no correlation between serum betatrophin levels and any indexs in T2 DM group. In HINS group, serum betatrophin levels were correlated with age, waist, and correlated with age,AST,TG,FINS levels in NC group.5.Serum 25-OH-VD levels correlated positively with HOMA-IR and negatively with age and serum betatrophin levels, had no correlation with other indexes. Conclusion 1.In the presence of hyperinsulinemia, serum betatrophin levels decreased and serum betatrophin levels increased in T2 DM patients of handicapped beta cell, suggested that the serum betatrophin levels correlated with insulin resistance and beta cell function, and betatrophin may be involved in the progression of diabetes.2.Age and serum 25-OH-VD levels were independent factors affecting serum betatrophin levels.
