Viburnum.a genus belonging to the family Adoxaceae(formerly Caprifoliaceae),consists of more than 230 species of shrubs or small trees.Viburnum fordiae Hance,widely distributed in China,belongs to the genus Viburnum.As a traditional Chinese medicine,it has been widely used for the treatment of rheumatoid arthritis,lymphadenitis,tumefaction,cough and infantile tantrum.32 compounds were isolated from V.fordiae Hance by using various chromatographic and recrystallized methods(silica gel,RP-C18,MCI GEL,Sephadex LH-20 column chromatography,PTLC and HPLC).Their structures including absolute configurations were determined by spectroscopic and chemical methods.Triterpenoids:uvaol(1),28-nor-urs-12-ene-3,β,17β-diol(2),3-O-isopropylidenyl-2α,3α,19α-trihydroxyurs-12-en-28-oic acid(3),erythrodiol(4),oleanolic acid(5),lupeol(6);monoterpenoids:megastigmadien-3,9-dione(7),loliolide(8),dehydrololiolide(9),2a-hydroxycineole(10);lignanoids:(+)-isolariciresinol(11),fordianole A(12),fordianole B(13),fordianole C(14);phenylpropanoids:umbelliferone(15),3-(4-hydroxy-3-methoxyphenyl)propane-1,2-diol(16),7,8-bis-O-isopropylidene-dihydroeugenol(17),4-(3,4-dihydroxyphenyl)-5-methyl-dihyro-furan-2-one(18),1-(4-hydroxy-3-methoxyphenyl)-1-methoxypropan-2-ol(19),coniferyl aldehyde(20),p-hydroxylcinnamaldehyde(21),(+)-2-hydroxy-1-(4-hydroxy-3-methoxypheny)propan-1-one(22);phenolic acids:syringaldehyde(23),protocatechuate(24),3,4-dihydroxybenzoic acid methyl ester(25),vanillin(26),p-hydroxybenzaldehyde(27),salicylic acid(28),hydroquinone(30);others:benzyl alcohol(29),1-methyl-cyclopentene(31),β-sitosterol(32).Among them,12-14,17 and 18 were new compounds,and compounds 2,3,7-11,16,19-29 and 31 were isolated from this genus for the first time.The anti-inflammatory and antioxidant activities of the three extracts(PE,EtOAc and n-Butanol)from V.fordiae and compounds 1-30,isolated from the PE extract,were evaluated in vitro.The PE partition had preferable anti-inflammatory and antioxidant activities.And 20,21,30 showed potent inhibitory activity on NO production in RAW264.7 cells stimulated by LPS.11-14,18,20,21,23,24,26,27,30 exhibited varying antioxidant activities through DPPH,ABTS free radical scavenging and FRAP assays.
