ObjectiveBy detecting the effect of Qishaofumai decoction on serum enzymes index.CD34+ischemic skeletal muscle capillary density(MVD),vascular endothelial growth factor(VEGF),and the histopathological test of limb ischemia rat,to discuss the Qishaofumai decoction,s effect on Rats Hind Linb ischemia model neovascularization and its possible mechanism,and also to provide basis diseases.MethodsUsed 90 SD rats with left lower limb artery ligation and branch enucleation to make the Hind Limb ischemia model.Divided them randomly into: model group.Qishaofumai decoction of high,medium and low dose group,and granulocyte colony-stimulating factor(rhG-CSF)group with 18 rats in each group.At the same time,set up a 18-rats sham-operated group,which only poerated the skin incision and sutures,but the femoral artery ligation.Then begin from the 3nd day after the surgery,give the rats medcine.The groups with Qishaofumai decoction of high,medium and low dose are in accordance with7.6 g/kg,3.8 g/kg,1.9 g/kg,respectively daily for lavage,lasting for 14 days.rhG-CSF group hypodermic 0.21 ml/100 g a day,lasting for 5 days.And the sham-operated water.Toop 6 rats from each group at 3 days,7 days and 14 days after operation,then detected them with automatic biochemical analyzer on peripheralblood creatine kinase(CK),lactate dehydrogenase(LDH),asperate aminotransferase(AST);also defected on thepercentage of CD34+ positive cells in serum with flow cytometry;Took the operation side of adductor muscle,and measured its density of VEGF and MVD by immunohistochemical method;Observed the pathological changes of gastrocnemius with hematoxylin-eosin(HE)and compared differences.Results1.MVD results: 14 days post operatively,the MVD of Qishofumai decoction of high,medium dose groups were singnificanthy higher than the model group and low dose group(P<0.01);and high-dose group,s MVD values higher than medium dose one.2.The enzyme test results: at 3 days after poeration,with the exception of sham-poerated group.Groups of LDH,CK and AST values were increased significantly(P<0.05 or P<0.01);at 7 days after operation,except for the sham group,each group,s LDH,CK and the AST values were decreased quickly than that of in the former 3 days,and it,s worthwhile to note that the declining datas of high,medium dose groups and the rhG-CSF group were the most obvious(P<0.01);at 14 days,each group,s LDH,CK and AST declined slowly.3.Proporation of CD34+ positive cells test resalts : at each time point postoperative,the percentages of CD34+cells positive of high Qishaofumai dose group were higher than those of the model group(P<0.01);At 7 days and 14 days after operation,the proportions of CD34+positive cells of high Qishaofumai dose group were significantly higher than that of the medium and low dose ones(P<0.01);At 7 days after peration,the proportions of positive cells of high,medium dose groups and rhG-CSF group increased clearly than those of in the former 3 days(P<0.01).After 14 days,all group,s CD34+ proprotion decreased(P<0.05 or P<0.01).4.VEGF protection expression: At each time point after operation,the VEGF density indexes of Qishaofumai high dose groups rose substantially(P<0.01)than those of the model group.7 days after surgery,in addition to sham-opreated group,the expression of each group increased;14 days after surgery,in addition to sham-operated group,every expression reduced.5.Pathological result: The cataplasia with high Qishaofumai dose was highter,and the number of the blood vessels among the muscle fibers increased evidently than that of the model one.ConclusionQishaofumai decoction can increase the ischemia skeletal muscle,s capillary density,promote the neovascularization,and it,s mechanism may have a link with the rising VEGF,mobilizing the marrow EPCs move to the ischemic tissue and promoting the regenerotton of the endothelial cells.
