This paper is funded by the forestry public welfare industry special "Curculigo,Millettia resources cultivation and undergrowth cultivation technology"(201304807)funding.To establish HPLC fingerprint for the identification and evaluation of Millettia and Curculigo.The samples were collected under by the natural wild conditions of the growth of Millettia and Curculigo orchid GAP planted,and then will be processed by ultrasonic extraction,solid phase extraction and high performance liquid chromatography(HPLC).The results were shown,get a total peak fingerprint.Millettia reverse phase chromatography conditions:Column:GL Science C8(460×250mm,3?m);The flow rate:0.6 ml/min;Column temperature:25?;Detection wavelength:260nm;Mobile phase:0.2%acetic acid-ddH2O:0.2%acetic acid-100%acetonitrile(A:B)ratio,gradient elution;after running time:5min,Curculigo chromatographic conditions:Column:GL Science Cs(460×250 mm,3?m);Mobile phase:A:0.1%phosphoric acid-ddH2O;B:0.1%phosphoric acid-100%acetonitrile;Flow rate:0.8ml/min;Detection wavelength:282nm;After running time:2min.The chromatographic fingerprints and standard fingerprints of the samples were analyzed and the chemical composition was determined.The main findings are as follows:(1)The contents of five Flavonoids in Millettia were determined by HPLC,the content of Isoliquiritigenin was about 3.45%in Guangxi Dagui Mountain from 50 to 60 years old Millettia,followed by Hezhou of Guangxi 5-6 years'Millettia,the content of Rutin in stem was about 1.73%,and that of Durumin in mature leaves of 5-6 years old Millettia in Hezhou of Guangxi was about 0.66%.Using EXCEL for data processing,combined with IBM SPSS Statistics software for Univariate Analysis of Variance and Difference Analysis,inferred the content of five flavonoids in 10 batches of Millettia was significantly different.Daidzein,Maison Isoflavones,Glycyrrhizin,Isocarizin,and Rutin were all good in linear relationship in the range of 0.002g-0.01g.(2)In the Millettia fingerprints' construction experiment,mainly to study Dense Beans,by HPLC experiment,I got the result:a total of 18 batches of test samples,and then use the "Chinese medicine chromatographic fingerprint similarity evaluation system 2.0" software analysis to obtain chromatographic fingerprints,through the optimization of the optimal chromatographic conditions to build the fingerprints can be a good reveal of its inherent characteristics,the separation effect and similarity of the chromatographic peaks were good.Construction of Chromatographic Fingerprints can identify genuine and obscure quality of the herbs.There are five common fingerprint peaks in the fingerprints of Millettia in this study,5 common fingerprint peaks were peak A:Protocatechuic acid,peak B:Epicatechin,peak C:Rutin,peak D:Thimerosulf-onate,peak E:Daidzein,this study is mainly based on Dense Beans,so it can be determined as a common peaks of Dense Beans fingerprints.And I distincted my research on the Millettia's genuine products and Millettia's obscure products of the 18 batches of Millettia.(3)Using EXCEL,HPLC experiment and"Chinese medicine chromatography fingerprint similarity evaluation system 2.0" software to analysis and processing data,I got the result that in the Curculigo fingerprints' construction experiment,there are 5 common fingerprint peaks in the fingerprints,peak E:fairy glycoside,other peaks have not yet been purchased to the corresponding standard samples,it does't determine which specific peak belongs to which material,but from Figure 4.2 can determine the four substances' content are high.Curculigo chromatogram construction can also be used for the genuine and obscure quality identification of the herbs.The content of fairy glycoside in the Chinese Curculigo from three different habitats of Guangxi,Sichuan and Yunnan was measured,Yunnan fairy glycoside's content in Chinese Curculigo is about 6.844%,Sichuan fairy glycoside's content in Chinese Curculigo is about 7.719%,Guangxi fairy glycoside's content in Chinese Curculigo is about 7.189%,the average content of fairy glycoside is about 7.251%.
