ObjectiveTo evaluate the role of chitosan in Vaseline gauze(VG)for wound healing in vitro and in vivo,we prepared a functional Chitosan-Vaseline gauze(CVG)and studied its cytotoxicity,antibacterial property,moisture retention and its effect on wound healing.MethodsHigh and low molecular weight chitosan were incorporated into sterile gauze to build a functional Chitosan gauze.Then dried Chitosan gauze was soaked in mixed Vaseline-paraffin oil to fabricate CVG.Scanning electron microscopy(SEM)and infrared(IR)spectroscopy were used to observe the microstructure of CVG.Characteristics of CVG,including cytotoxicity(Cell Counting Kit-8 assay),antibacterial activity(inhibition zone test)and moisture retention(digital comeometer)were tested.Histopathological measurement was done after HE staining,CD31 Immunohistochemical(IHC)staining and Masson staining.ResultsThe results of SEM and IR spectroscopy demonstrated the successful incorporation of chitosan into VG Compared with VG.CVG showed better antibacterial properties and moisture-retention with little cytotoxicity to NIH3T3 cells.Wounds dressed with CVG showed a significant increase in wound healing rate and shorter epithelialization time(14.83 ± 0.75 days versus 18.50 ± 0.55 days,P = 0.00).CVG also effectively promoted horizontally-oriented collagen and faster angiogenesis.ConclusionCompared to VG,chitosan in CVG offers good antibacterial,moist environment and promotes wound healing evidently with little cytotoxicity to NIH3T3 cells.
