Clinical Observation Of Low-dose Letrozole Combined With Tadalafil In The Treatment Of Hyperestrogenic Erectile Dysfunction

Objective:To evaluate the safety and efficacy of low-dose Letrozole combined with tadalafil in the treatment of hyperestrogenic erectile dysfunction.Methods:This study involved 55 erectile dysfunction outpatients with hyperestrogenism enrolled in Andrology clinic from January 2015 to July 2016.55 patients were divided into the Observation group(n=28)and the Control group(n=27)depengding the order of treatment.All patients were given orally 5mg Tadalafil per night for 12 weeks.The Observation group were given orally 2.5mg Letrozole tablets additionally before breakfast on Monday and Thursday,while The Control group was only given orally 5mg Tadalafil per night.The effect of treatment was evaluated by the International Erectile Function Questionnaire(IIEF-5),the weekly frequency of sexual intercourse,Sexual Encounter Profile Question3(SEP3: whether you successfully completed sexual intercourse),the E2 level,while the adverse effects of treatment were monitored to evaluate the safety? Results:There was no significant difference between the two group in general characteristics before treatment,and after 12 weeks of treatment:1.The scores of IIEF-5 and the success rate of sexual intercourse were significantly improved in the two groups(p <0.05);besides,they were higher in the Letrozole group than those in the Control group(p <0.05).2.The weekly frequency of sexual intercourse in the Letrozole group was significantly improved,and it was higher than that in the control group(p <0.05).Before and after treatment,there was no significant change in the weekly frequency of sexual intercourse of the Control group(P> 0.05).3.The E2 of the Letrozole group was significantly lower than that of beforetreatment or the control group(p <0.05);there were no significant change in TT level of the two groups(p> 0.05).4.Serious incidences in two groups were not observed,such as liver and kidney dysfunction,cardiovascular events,water and sodium retention and so on.There were no significant difference in the incidence of adverse events of two group(17.9% s22.2%,p<0.05).Conclusion:1.Low-dose Tadalafil in the treatment of hyperestrogenic erectile dysfunction can improve symptoms,but the efficacy is poor.2.The treatment of low-dose of letrozole combined with tadalafil in patients with hyperestrogenic ED can decrease estrogen levels,restore erectile function,improve sexual desire,and have little adverse events,it can be thought that the treatment of letrozole combined with tadalafil is safe and effective.
