ObjectiveAs the change of awareness about treating disease,many scholars are attracted by the idea which applying immunostimulants to regulate the immune function.Immunostimulants are often used in the cancer treatment,they can enhance immune response when they individually or simultaneously used with antigen.The drugs used in cancer treatment as Cisplatin,Cyclophosph-amide,Mechlorethamine are lack of sufficient selectivity on tumor cells,as a result,they also attack the normal cells,which have serious impact on quality of patients' life.Long-term use of chemotherapy drugs can reduce the immune regulation function,their adverse effect include bone marrow suppression,leucopenia,thrombocytopenia,and so on.Scholars based on the research of the traditional Chinese medicine,try to find out safe,reliable herbal natural medicines of increasing immune function.Also these herbal natural medicines are expected to improve the inhibitory effect of immune function induced by the chemotherapy drugs.This experiment,through in-vitro spleen lymphocyte proliferation and NK cell activity experimental screening the optimum combination of Composite Rhodiola rosea(CRr).Moreover,Fermented Composite Rhodiola rosea(FCRr)and non-fermented Composite Rhodiola rosea(NCRr)carry on the contrast experiments to explore the increasing effect of immune response and its mechanism.Methods1.According to active yeast fermentation condition,extracting separately Rhodiola rosea and Panax ginseng.By the factorial design,according to CRr extracts in-vitro measurement on immune function,select the optimal combination of CRr.2.According to the serum pharmacological experiments,taking nine rats,according to factorial design test combining nine different CRr groups,instragastric administrating these liquid continuously five days,2 hour after last delivery,then separating the collect serum in rats.According to MTT method,observe in-vitro spleen lymphocyte proliferation and NK cell activity.3.Increasing immune function in-vivo experiments,randomly divided into six groups;normal control group,immunosuppressive model group,and two groups treated respectively with low and high dose of FCRr,and two groups treated respectively with low and high dose of NCRr.The immunosuppressive mice model was constructed by intraperitoneal injection with 100mg/kg BW cyclophosphamide for three times,then FCRr and NCRr were given by intragastric administration for 15 days and the number of white blood cell and spleen lymphocyte proliferation and NK cell activity and the concentration of IL-2,IFN-?,TNF-a were observed.Results1.CRr can significantly promote T and B lymphocyte proliferation(P<0.01).Fermented and non-fermented 0.6g/kg Rhodiola rosea and 1g/kg Panax ginseng might significantly promote T and B lymphocyte proliferation and significantly increase NK cell activities(P<0.01).Fermented and non-fermented 0.3g/kg Rhodiola rosea and 0.5g/kg Panax ginseng might significantly promote T and B lymphocyte proliferation and significantly increase NK cell activities(P<0.01).2.The immunosuppressive mice model was constructed by cyclophosphamide,400mg/kg and 800mg/kg FCRr and NCRr might significantly increase the number of white blood cell(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 80000mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly increase the number of white blood cell(P<0.01),in immunosuppressed mice.3.400mg/kg and 800mg/kg FCRr and NCRr might significantly promote T and B lymphocyte proliferation(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 800mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly promote T and B lymphocyte proliferation(P<0.01),in immunosuppressed mice.4.400mg/kg and 800mg/kg FCRr and NCRr might significantly increase the NK cell activities(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 800mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly increase the NK cell activities(P<0.01),in immunosuppressed mice.5.Cyclophosphamide might significantly decrease the concentration of IL-2(P<0.01),400mg/kg and 800mg/kg FCRr and NCRr might significantly increase the concentration of IL-2(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 800mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly increase the concentration of IL-2(P<0.01),in immunosuppressed mice.6.Cyclophosphamide might significantly decrease the concentration of IFN-?(P<0.01),400mg/kg and 800mg/kg FCRr and NCRr might significantly increase the concentration of IFN-?(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 800mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly increase the concentration of IFN-y(P<0.01),in immunosuppressed mice.7.Cyclophosphamide might significantly decrease the concentration of TNF-a(P<0.01),400mg/kg and 800mg/kg FCRr and NCRr might significantly increase the concentration of TNF-a(P<0.01),in immunosuppressed mice.Compared with 400mg/kg and 800mg/kg NCRr,400mg/kg and 800mg/kg FCRr might significantly increase the concentration of TNF-a(P<0.01),in immunosuppressed mice.Conclusion1.The concentration of the optimum combination of Rhodiola rosea and Panax ginseng in in-vitro study as below,High doses are 0.6g/kg Rhodiola rosea and 1g/kg Panax ginseng,low doses are 0.3g/kg Rhodiola rosea and 0.5g/kg Panax ginseng.2.The concentration of the optimum combination of Rhodiola rosea and Panax ginseng in in-vivo study as below,FCRr and NCRr high doses are 800mg/kg,FCRr and NCRr low doses are 400mg/kg.3.Fermenting Rhodiola rosea and Panax ginseng with active yeast might significantly increase the immune activity,moreover this method is convenient to use.
