Objective: Laryngeal squamous cell carcinoma is a common malignancy in head and neck region,accounting for 13.9% of head and neck tumors.At present,laryngeal cancer is studied extensively,and the research on protein,pathway and various aspects is concentrated,Laryngeal cancer through which a specific mechanism of the main role,there is no exact expression of the occurrence of laryngeal cancer.The development of multiple factors or pathways caused by the role of the recent signal pathway research has become a hot spots for the laryngeal cancer occurrence and development study.To find a new therapeutic target for the eatment of laryngeal cancer and provide reference indicators in prognosis of laryngeal cancer.The signal transduction pathway is one of the most typical signal transduction pathways closely related to tumor.The main expression pattern is that when the wnt signal is activated by the initiating factor,(TCF / LEF)in the nucleus,resulting in the growth of the cells,which resulted in the inhibition of ?-catenin phosphorylation and the degradation of ?-catenin.Dvl and Wnt2 are the main members of the wnt signaling pathway,located upstream of the whole signal transduction pathway.The expression of the two proteins in the pathway is the key link of the signal transduction pathway.The expression of the two proteins is abnormal,which leads to ?-catenin in the cytoplasm level increased accumulation of nuclei,and transcription factors interact,induced the transcription of target genes,refer to the previous literature Wnt / ?-catenin pathway dvl and wnt2 in esophageal cancer,non-cell lung cancer,glioma,Breast cancer,colon cancer were abnormal expression.In this study,the expression of Dv1 protein and Wnt2 in laryngeal squamous cell carcinoma and normal peripheral tissues were detected by immunohistochemistry.The effects of two proteins on the age,location, clinical stage,metastasis,And to explore the pathogenesis of Dvl and Wnt2 in laryngeal squamous cell carcinoma,and to provide a new way to find a new therapeutic approach.Method:1 study object1.1 The collection of paraffin specimens selected from the Fourth Hospital of Hebei Medical University ENT-Head and Neck Surgery in October 2014 to November 2016 in Hebei Medical University Hospital,the fourth hospital in the laryngeal tumor patients,of which the experimental group of 48 cases,Pathologically identified patients with squamous cell carcinoma,confirmed by medical records confirmed that this group of patients were selected before surgery were not put,chemotherapy and other treatment,aged 43 to 71 years,mean 58 years old,the control group of 24 cases of laryngeal cancer patients Of the adjacent normal mucosal tissue,away from the primary site of cancer0.5 cm.The expression of Dv1 and Wnt2 in the experimental group and the control group were detected by immunohistochemistry(IHC)SP method.1.2 All the experimental data were analyzed by SPSS21.0 software.?2 test,P<0.05 was considered statistically significance.Result:1 The expression of DvlThe positive expression of Dvl protein was brown,mainly located in the cytoplasm,mostly diffuse expression.The positive expression rate of Dvl expression in laryngeal squamous cell carcinoma(79.2%)was significantly higher than that in para-carcinoma normal tissues(33.3%),and the difference was statistically significant(P<0.05).1.1 The positive expression rates of Dvl in poorly differentiated and welldifferentiated pathologic types were 89.4% and 65.5% respectively,which indicated that the expression of Dvl in cancer tissues increased with the degree of differentiation,and the difference was statistically significant(P<0.05).1.2 The expression of Dvl protein was correlated with the clinical stage and the positive expression of ?+?(57.1%)was significantly lower than that of? + ?(85.5%),and the difference was statistically significant(P<0.05).1.3 The expression of Dvl protein was significantly higher in patients with lymph node metastasis(84.6%)than that without lymph node metastasis(63.6%),and the difference was statistically significant(P<0.05).1.4 The expression of Dvl protein in laryngeal squamous cell carcinoma was not significantly correlated with age and sex(P> 0.05).2 The expression of Wnt22.1 Wnt2 positive substances mainly in the cytoplasm,yellow or brown particles.The positive expression of Wnt2 in laryngeal squamous cell carcinoma(75.0%)was significantly higher than that in para-carcinoma normal tissues(45.8%).There was significant difference between the two groups(P<0.05).2.2 The expression of Wnt2 was 84.2% and 58.60% in poorly differentiated and well-differentiated laryngeal squamous cell carcinoma,respectively.The lower the differentiation degree of wnt2 was higher than positive expression rate,The difference was statistically significant(P<0.05).2.3 The expression of Wnt2 was correlated with clinical stage.The expression rate of Wnt2 was significantly higher than that of stage ? and ?(60.7%).The positive expression rate of Wnt2 was significantly higher than that of stage ? and ?,The difference was statistically significant(P<0.05).2.4 The expression of Wnt2 in laryngeal carcinoma with lymph node metastasis(84.6%)was significantly higher than that in lymph node metastasis(59.1%),and the difference was statistically significant(P<0.05).3 Relationships of Dvl and Wnt2The relationships between Dvl and Wnt2 expression in laryngeal squamous cell carcinoma was positively relationships(r = 0.537,P<0.001).Conclusions:1 The expression of Dvl and Wnt2 in laryngeal squamous cell carcinoma was significantly higher than that in para-carcinoma normal tissues,suggesting that both of them may be involved in the development and progression of laryngeal carcinoma tissue malignancy and lymph node metastasis.2 The expression of Dvl in laryngeal squamous cell carcinoma was significantly higher than that in clinical stage,differentiation and lymph node metastasis,suggesting that the high expression of Dv1 protein may play an important role in the development,invasion and metastasis of laryngeal carcinoma.3 Wnt2 has a high expression in the clinical stage,differentiation and lymph node metastasis of laryngeal squamous cell carcinoma,suggesting that Wnt2 protein may promote the development of laryngeal carcinoma and may be a characteristic marker of laryngeal squamous cell carcinoma.4 Dvl protein and Wnt2 protein expression in laryngeal cancer have a positive correlation between the two may be through a mechanism in the occurrence and development of laryngeal carcinoma play a role in the targeting of laryngeal cancer provides a new idea,But whether it can become a new treatment of laryngeal cancer need further clinical validation.
